摘 要:目的 研究海南省屯昌蚊种多样性,并进行系统发育分析。方法 2019年8月以人诱法采集海南省屯昌县蚊虫标本,HotShot法提取蚊虫DNA,PCR扩增ITS2基因片段,测序,经BLAST比对明确蚊种,用Mega X软件进行系统发育分析。结果 采集到蚊虫43只,其中18只雌蚊成功测序,分别为骚扰阿蚊6只,占33.33%;白纹伊蚊5只,占27.77%;白雪库蚊和淡色库蚊各2只,河滨伊蚊1只,未定种库蚊2只。ITS2基因序列变异位点比例为54.09%,具有较高的变异性,不存在碱基替换饱和。系统发育分析显示,海南省屯昌的不同蚊种分别与美国、斯里兰卡、日本、芬兰、希腊、以色列以及中国的重庆、上海、福建的相应蚊种系统发育关系相近,预测与蚊虫迁徙、扩张有关。结论 研究蚊多样性和监测进化动态,是预警和防控蚊媒病传播的基础。Objective To study the diversity and phylogenetic relationship of mosquito species in Tunchang,Hainan province. Methods The mosquitoes were collected by using human-landing catch(HLC) method in August 2019 in Tunchang county,Hainan province. Genomic DNA was extracted by HotShot method and the ITS2 gene fragment was amplified by PCR and sequencing. The mosquito species were identified in the BLAST alignment. The phylogenetic analysis was performed with MEGAX software. Results A total of 43 mosquitoes had been captured,among them,18 mosquitoes were successfully sequenced,including 6 Armigeres subalbatus(33.33%),5 Aedes albopictus(27.77%),2 Culex gelidus(11.11%),2 Culex quinquefasciatus(11.11%),1 Aedes riversi(5.56%) and 2 Culex spp.. The percentage of mutation sites in ITS2 gene sequence was 54.09%,with high variability and little substitution saturation. Phylogenetic analysis showed that the phylogenetic relationships of different mosquito species in Tunchang were similar to those in USA,SriLanka,Japan,Finland,Greece,Israel,as well as with those in Chongqing,Shanghai and Fujian of China,respectively,related to the migration and expansion of mosquitoes. Conclusion Studies on mosquito diversity and monitoring of evolutionary dynamics are fundamental for early warning and control of mosquito-borne disease transmission.
