摘  要:目的研究五味子提取物对大鼠肝脏中细胞色素P450酶(CYP450酶)的影响。方法采用高效液相色谱串联质谱(high performance liquid chromatography tandem mass spectrometry,HPLC-MS)分析技术建立稳定的探针底物色谱分析法,结合Cocktail分析单次给予五味子提取物对大鼠体内CYP2D6、CYP2C19、CYP2C8、CYP2A6、CYP2B6、CYP2C196种CYP450亚型酶活性的影响。结果五味子提取物对大鼠肝脏中CYP2D6、CYP2C19和CYP2C8的活性具有显著的抑制作用,而对CYP2A6、CYP2B6和CYP2C19的活性具有显著的诱导作用(P<0.05)。结论用HPLC-MS结合Cocktail探针药物法研究五味子提取物对大鼠CYP450酶影响的准确性及可靠性较强,五味子对CYP450酶多个亚型的不同调控作用可能是其发挥降酶保肝作用的重要机制。Objective To study the effect of Schisandra chinensis extract on 6 common and important CYP450 enzymes in rat liver.Methods HPLC-MS was used to establish a stable probe substrate chromatography method, and cocktail analysis was used to evaluate the activities of 6 CYP450 isoforms CYP2 D6,CYP2 C19,Cyp2 C8,CYP2 A6,CYP2 B6 and CYP2 C19 in rats.Results Schisandra chinensis extract could significantly inhibit the activities of CYP2 D6,CYP2 C19 and CYP2 C8,and induce the activities of CYP2 A6,CYP2 B6 and CYP2 C19(P<0.05).Conclusion The method established in this study is reliable and accurate.The different regulatory effects of Schisandra chinensis on CYP450 isoforms may be one of the important mechanisms of Schisandra chinensis on reducing enzyme and protecting liver.
