摘  要:目的观察白藜芦醇对IgA肾病大鼠肾功能的影响以及对炎性反应的作用。方法将48只SD大鼠分为空白对照组、IgA肾病模型组、白藜芦醇组和氯沙坦组。除空白对照组外,其余3组大鼠构建IgA肾病模型,白藜芦醇组给予20 mg·kg^(-1)·d^(-1)白藜芦醇,氯沙坦组给予16.8 mg·kg^(-1)·d^(-1)氯沙坦,空白对照组和IgA肾病模型组给予等体积生理盐水,灌胃8周。检测4组大鼠血清白细胞介素(interleukin,IL)-6、IL-1β和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的水平,检测血肌酐、尿素氮和24 h尿蛋白的含量,观察大鼠肾组织IgA免疫荧光和病理学变化,用蛋白质免疫印迹(Western blot)检测大鼠肾组织Notch信号通路的变化。结果IgA肾病模型组大鼠血清中IL-6、IL-1β、TNF-α、肌酐、尿胆素、24 h尿蛋白、IgA沉淀、Notch1和Jagged1蛋白的表达量均高于空白对照组(P<0.05),白藜芦醇组和氯沙坦组大鼠血清中IL-6、IL-1β、TNF-α、肌酐、尿胆素、24 h尿蛋白、IgA沉淀、Notch1和Jagged1蛋白的表达量均显著低于IgA肾病模型组(P<0.05)。白藜芦醇组和氯沙坦组上述指标的差异无统计学意义(P>0.05)。结论白藜芦醇可保护IgA肾病大鼠的肾功能,减轻炎性反应和IgA沉淀,并抑制Notch信号通路的转导。Objective To observe the effect of resveratrol on renal function of rats with IgA nephropathy and its effect on inflammatory response.Methods 48 SD rats were divided into blank control group, IgA nephropathy model group, resveratrol group and losartan group.Except for the blank control group, the other 3 groups were used to establish IgA nephropathy models.The resveratrol group are given resveratrol 20 mg·kg^(-1)·d^(-1) and the losartan group are given losartan 16.8 mg·kg^(-1)·d^(-1).The blank control group and IgA nephropathy model group were given an equal volume of normal saline.The gastric gavage cycle was 8 weeks.The levels of interleukin(IL)-6,IL-1β and tumor necrosis factor-α(TNF-α) in serum, and the levels of blood creatinine and urea nitrogen were detected.The IgA immunofluorescence and pathological changes were observed in rat kidney tissue.And the changes of signal pathway in rat kidney tissue were detected by Western blot.Results The serum levels of IL-6,IL-1β,TNF-α,creatinine, urobilin, 24-hour urine protein, IgA precipitation, Notch1 and Jagged1 protein expression in the IgA nephropathy model group were higher than those in the blank control group(P<0.05).The serum levels of IL-6,IL-1β,TNF-α,creatinine, urobilin, 24-hour urine protein, IgA precipitation, Notch1 and Jagged1 protein expression in the resveratrol group and losartan group were significantly lower than those in IgA nephropathy model group(P<0.05).There was no statistically significant difference in the above indicators between the resveratrol group and the losartan group(P>0.05).Conclusion Resveratrol could protect the renal function of rats with IgA nephropathy, reduce the inflammation and IgA precipitation, and inhibit the conduction of Notch signaling pathway.
