摘 要:目的研究菊苣酸(chicory acid,CA)对内毒素诱导的大鼠急性肺损伤(acute lung injury,ALI)的保护作用及机制。方法将90只雄性SD大鼠随机分为6组:假手术组、CA组、高剂量CA组、中剂量CA组、低剂量CA组以及LPS组,每组15只。假手术组灌胃给予生理盐水(5 mL/kg)1 h后,腹腔注射生理盐水;CA组将CA(40 mg/kg)溶解于生理盐水灌胃1 h后,腹腔注射生理盐水;高、中、低剂量CA组分别灌胃CA 40 mg/kg、20 mg/kg、10 mg/kg后腹腔注射LPS(10 mg/kg);LPS组灌胃给予生理盐水(5 mL/kg)1 h后,腹腔内注射LPS(10 mg/kg)。苏木精-伊红(HE)染色组织切片评估肺损伤;测定肺湿干重比、支气管肺泡灌洗液(BALF)中的蛋白质含量和肺组织中的Evans蓝(EB)含量,以EB含量与湿肺重之比评估肺微血管通透性;免疫组织染色法检测骨髓过氧化物酶(MPO)及Western blotting法测定紧密连接蛋白Occludin、ZO-1、JAM-1的表达水平。结果HE染色结果显示,LPS组肺间质组织明显水肿增厚,CA处理组减轻了LPS诱导的肺水肿;免疫组化结果显示,各CA处理组的MPO阳性染色有明显下降;与LPS组相比,CA 20 mg/kg和40 mg/kg组的肺湿/干重比、EB外渗、BALF蛋白浓度和BALF总细胞数均显著降低;在CA 20 mg/kg和40 mg/kg剂量组中,肺组织紧密连接蛋白ZO-1、Occludin和JAM-1的表达量显著高于LPS组。结论CA对LPS诱导的大鼠ALI具有保护作用,其机制可能与CA可抑制肺组织微血管屏障的破坏有关。Objective To study the protective effect of chicory acid(CA)on endotoxin-induced acute lung injury(ALI)in rats.Methods Ninety male SD rats were randomized into six groups,sham,CA,high dose CA,medium dose CA,low dose CA,and LPS,with 15 rats in each group.Sham operation group was given normal saline(5 mL/kg)by intragastric administration for 1 hour,then intraperitoneal injection of normal saline.In the CA group,CA(40 mg/kg)was dissolved in normal saline for 1 hour,followed by intraperitoneal injection of normal saline.High,medium and low dose CA groups were intragastric administration of CA40 mg/kg,20 mg/kg and 10 mg/kg,respectively,followed by intraperitoneal injection of LPS(10 mg/kg).LPS group was given normal saline(5 mL/kg)by intragastric administration for 1 hour,then intraperitoneal injection of LPS(10 mg/kg).Lung injury was evaluated using hematoxylin and eosin staining techniques.Lung wet-dry weight ratio,protein content in bronchoalveolar lavage fluid(BALF)and Evans blue(EB)content in lung tissue were determined,and the ratio of EB content to wet lung weight was used to evaluate pulmonary microvascular permeability.Myeloperoxidase(MPO)expression levels was detected by immunohistochemical staining.Western blotting was used to measure the expression level changes of tight junction proteins,including zonula occludens ZO-1,Occludin and junctional adhesion molecule JAM-1.Results HE staining showed that the pulmonary interstitial tissue was significantly thickened in the LPS group,and the CA-treated group alleviated the pulmonary edema induced in the LPS.Immunohistochemistry results showed a significant decrease in positive MPO staining in each CA treatment group.The lung wet/dry weight ratio,EB extravasation,BALF protein concentration,and total BALF cell number were significantly lower in the CA 20 mg/kg and 40 mg/kg groups when compared with the LPS group.Expression of the lung tissue tight junction proteins ZO-1,Occludin,and JAM-1 was significantly higher in the CA 20 mg/kg and 40 mg/kg doses than those in t
