摘 要:目的 利用网络药理学探讨黄芩治疗慢性乙型病毒性肝炎(CHB)的主要成分、作用靶点及通路。方法 通过中药系统药理学数据库与分析平台(TCMSP),在类药五原则(rule of five)的基础上按照生物利用度(OB)>30%,类药性(DL)>0.18的标准筛选出黄芩活性药物成分并下载其靶点信息,利用GeneCards和Omim数据库统计慢性乙型病毒性肝炎靶点,使用Uniport数据库对上述药物和疾病的共有靶点信息进行靶基因标注,利用Cytoscape软件制作“药物成分-疾病-靶点”关系图,通过String平台绘制蛋白互作网络(PPI),以Bioconductor平台为背景使用R语言程序进行GO富集分析和KEGG富集分析。结果 共筛选出活性成分32种。药物和疾病的共有靶基因85个。PPI网络核心基因包括AR,AKT1,IL6等共计15个。GO富集分析得到生物过程132条,如键合DNA转录活化剂活性,泛素样蛋白连接酶结合等。KEGG富集分析得到包括内分泌抵抗通路,细胞凋亡通路,甲型流感通路等124条信号通路。结论 黄芩对慢性乙型病毒性肝炎具有多基因、多靶点、多通道的作用特点,为后续筛选药物有效成分及中医新药研发提供了一定的设计思路和理论支持。Objective The main targets and pathways of traditional Chinese medicine Scutellaria baicalensis Georgi in the treatment of chronic viral hepatitis B(CHB)were discussed by using the research methods of systematic pharmacology and related databases.Methods The active components of Scutellaria baicalensis Georgi were screened and the target information was downloaded based on the rule of five,OB>30% and DL>0.18,by using the database and analysis platform of Systems pharmacology,the Gene Cards and Omim databases were used to count the targets of chronic viral hepatitis B,the Uniport database was used to annotate the target genes of these drugs and diseases,and the Cytoscape was used to map“drug composition-disease-gene”relationship,drawing protein-prvtein interaction(PPI)by String platform,using R language program to do GO(gene ontology)enrichment analysis and KEGG(Kyoto encyclopedia of genes and genomes)enrichment analysis on the background of Bioconductor platform.Results A total of 32 active components were screened out.There were 85 common target genes for drugs and diseases.There were 15 core genes in the PPI network,including Ar,AKT1,IL6,and so on.GO enrichment analysis resulted in 132 biological processes,such as binding DNA transcriptional activator activity,ubiquitin-like protein ligase binding,and so on.KEGG enrichment analysis revealed 124 signal pathways including endocrine resistance pathway,apoptosis pathway and influenza a pathway,and so on.Conclusion This study shows that Scutellaria baicalensis Georgi has multi-gene,multi-target,and multi-channel effects on chronic viral hepatitis B,which provides some design ideas and theoretical support for the following screening of effective components and the research and development of new Chinese medicine.
