摘 要:研究从患烂鳃病的扁吻鱼及其水样中分离到5株优势菌株,观察优势菌株的菌落形态及生长曲线,提取菌株16S rDNA进行分子生物学鉴定,并采用水产生产实践中常用抗生素对菌株进行药敏试验,同时探讨了培养基、培养时间、培养温度3个因素对菌株药敏性的影响。结果表明:编号33、37、38的菌株属于柠檬酸杆菌属(Citrobacter sp.),编号35、36的菌株属于气单孢菌属(Aeromonas sp.);生长曲线测定表明,5株细菌用MH培养基在18℃下培养时,0~18 h均处于适应期,33、37和38号菌株在培养28~32 h进入衰亡期,35和36号菌株在培养22~26 h进入衰亡期;综合培养基、培养时间、培养温度3个因素,最终判定,对土霉素敏感的菌株为33和38号,对恩诺沙星敏感的菌株为35和38号,对氟苯尼考敏感的菌株为33、35、36和38号,对多西环素敏感得菌株为33和38号,5个菌株对新霉素和甲氧苄啶均不敏感;培养时间对药敏试验结果的影响较小,而培养基和培养温度对药敏试验结果的影响较大。In this study,five dominant strains were isolated respectively from Aspiorhynchus laticeps individuals died of gill-rot disease and aquacultural water samples.They were cultured to observe the colonial morphology and growth curve,and then were conducted 16S rDNA sequence identification.Furthermore,the drug-sensitive test of these five strains was performed with ordinary antibiotics to test the influence of medium,cultural duration and cultural temperature on the drug sensitivity of the strains.The molecular identification results showed that strains 33,37 and 38 belonged to Citrobacter sp.,and strains 35 and 36 belonged to Aeromonas sp.The growth curve showed that all five strains were in the adaptation stage during 0-18 h culture in MH at 18℃,and then strains 33,37 and 38 entered the decline stage after 28-32 h culture,strains 35 and 36 entered the decline stage after 22-26 h culture.The drug-sensitive test results showed that strains 33 and 38 were sensitive to terramycin and doxycycline,strains 35 and 38 were sensitive to enrofloxacin,strains 33,35,36 and 38 were sensitive to florfenicol;all five strains were not sensitive to neomycin and trimethoprim;cultural duration had little influence,while cultural medium and temperature had large influence on drug sensitivity.
