摘　　要：目的探索ZWINT在黑色素瘤组织及细胞中的表达水平,及其对肿瘤细胞凋亡及迁移能力的影响。方法采用数据库分析及实时定量聚合酶链式反应检测ZWINT在黑色素瘤组织及细胞中的表达水平;通过数据库进一步分析ZWINT表达与黑色素瘤病理分期及预后的相关性;而后构建慢病毒干涉ZWINT表达;最后分别通过凋亡实验及Celigo划痕实验检测ZWINT对A375凋亡及迁移能力的影响。结果ZWINT在黑色素瘤组织及细胞中表达水平显著高于正常对照,高表达的ZWINT与黑色素瘤转移期及预后不良相关。敲降ZWINT可促进黑色素瘤细胞凋亡并抑制肿瘤细胞迁移能力。结论ZWINT在黑色素瘤组织及细胞内高表达并通过抑制凋亡及促进迁移能力发挥促癌作用。Objective To explore the expression level of ZWINT in melanoma tissues and cells and its effect on the apoptosis and migration of tumor cells.Methods The expression levels of ZWINT in melanoma tissues were analyzed by database and the expression levels of ZWINT in melanoma cells were detected by real-time quantitative polymerase chain reaction(RT-qPCR).The correlation between ZWINT expression and the pathological stage or prognosis of melanoma was further analyzed by database.Then lentivirus was constructed to knockdown ZWINT expression.Finally,the effects of ZWINT on the apoptosis or migration of A375 cells were detected by apoptosis test and celigo scratch test respectively.Results The expression level of ZWINT in melanoma tissues and cells was significantly higher than that of normal controls,and the highly expressed ZWINT was associated with the metastatic stage and poor prognosis of melanoma.ZWINT knockdown could promote the apoptosis of melanoma cells but inhibit the migration of tumor cells.Conclusion ZWINT is highly expressed in melanoma tissues and cells and plays an oncogenic role by inhibiting apoptosis and promoting migration of melanoma cells.
