摘 要:目的:研究益气温阳护卫汤对哮喘大鼠的治疗作用及其对肠道菌群的影响。方法:SPF级雄性SD大鼠24只,随机分为对照组、模型组、地塞米松组、益气温阳护卫汤组,每组6只。构建哮喘大鼠模型,药物干预4周后进行肺组织病理HE染色,16SrRNA测序进行菌群结构组成和多样性分析。结果:肺组织病理:对照组肺组织未见炎性病理改变;模型组可见典型哮喘病理表现;地塞米松和益气温阳护卫汤干预后肺组织病理改变得到明显改善。α多样性分析显示:模型组菌群丰度与多样性较对照组均降低,各药物组干预后菌群丰度和多样性均升高。在菌群门水平上,与对照组比较,模型组厚壁菌门比例降低;与模型组比较,益气温阳护卫汤组变形菌门和放线菌门比例升高。在菌群属水平上,与对照组比较,模型组乳酸杆菌属比例降低,益气温阳护卫汤干预后乳酸杆菌属比例略升高。LEfSe分析表明益气温阳护卫汤组肠道菌群的优势微生物主要集中在变形菌门和放线菌门。结论:益气温阳护卫汤能明显改善哮喘大鼠气道炎症,且对肠道菌群有一定的影响,该方药可能通过调节肠道菌群多样性和结构,发挥治疗哮喘的作用。Objective:To study the therapeutic effect of Yiqi Wenyang Huwei Decoction on asthmatic rats and its influence on the intestinal flora.Methods:Twenty-four SPF male SD rats were randomly divided into control group(A),model group(B),dexamethasone group(C),and Yiqi Wenyang Huwei group(YQWY),each with 6 rats.A rat model of asthma was constructed.After 4 weeks of drug intervention,pathological HE staining of lung tissue was performed,and 16SrRNA sequencing was performed to analyze the composition and diversity of bacterial flora.Results:Lung tissue pathology:no inflammatory pathological changes were seen in the lung tissue of the control group;Typical asthma pathological manifestations were seen in the model group;The pathological changes of lung tissue were significantly improved after the intervention of dexamethasone and Yiqi Wenyang Huwei Decoction.αdiversity analysis showed that the bacterial abundance and diversity of the model group were lower than those of the control group,and the abundance and diversity of the bacterial community increased after intervention in each drug group.At the level of phylum,compared with the control group,the proportion of Firmicutes in the model group decreased;Compared with the model group,the proportions of Proteobacteria and Actinomycetes increased in the YQWY group.At the level of genus,compared with the control group,the proportion of Lactobacillus in the model group decreased,and the proportion of Lactobacillus increased slightly after the intervention of Yiqi Wenyang Huwei Decoction.LEfSe analysis showed that the dominant microorganisms in the intestinal flora of the YQWY group were mainly concentrated in the Proteobacteria and Actinomycetes.Conclusion:Yiqi Wenyang Huwei Decoction can significantly improve airway inflammation in asthmatic rats,and has a certain effect on the intestinal flora.This prescription may play a role in treating asthma by regulating the diversity and structure of intestinal flora.
