摘  要:目的:基于Treg/Th17细胞及相关细胞因子探讨鹿鹅鼻炎方治疗过敏性鼻炎(AR)大鼠的作用机制。方法:45只AR大鼠随机分为空白组,模型组,西药组,低、高剂量中药组。观察大鼠鼻黏膜病理改变,检测鼻黏膜白细胞介素(IL)-17、IL-10表达,RT-PCR检测Foxp3 mRNA、RORγt mRNA的表达,ELISA检测大鼠血清IL-17、IL-10,TGF-β1的浓度。结果:与空白组比较,模型组大鼠鼻黏膜连续性中断,结构破坏,纤毛脱落,杯状细胞增生;药物干预组病理改变较模型组均有不同程度的减轻。与空白组比较,模型组大鼠鼻黏膜IL-10、Foxp3mRNA及血清中IL-10、TGF-β1含量显著降低(P<0.05),而大鼠鼻黏膜IL-17、RORγt mRNA、RORγt mRNA/Foxp3 mRNA及血清中IL-17含量显著升高(P<0.05)。与模型组比较,经药物干预后大鼠鼻黏膜IL-10、Foxp3mRNA及大鼠血清IL-10、TGF-β1均有不同程度的升高(P<0.05),鼻黏膜RORγt mRNA、RORγt mRNA/Foxp3mRNA及血清IL-17水平显著降低(P<0.05)。西药组、高剂量中药组大鼠鼻黏膜IL-17表达量较模型组显著下降(P<0.05)。结论:鹿鹅鼻炎方可改善大鼠鼻部过敏症状,通过调节体内Th17/Treg平衡而调节机体免疫状态,最终实现控制炎症反应达到治疗的目的。Obiective:To investigate the effects of Lu’e Biyan Formula on allergic rhinitis(AR)rats based on Treg/Th17 cells and related cytokines.Methods:Forty-five AR rats were randomly divided into blank group,model group,western medicine group,low-dose Chinese medicine group and high-dose Chinese medicine group.Pathological changes of nasal mucosa in rats were observed.The expression of IL-17 and IL-10 in the nasal mucosa of rats were measured.The mRNA expressions of Foxp3 and RORγt were detected by RT-PCR.The concentrations of IL-17,IL-10 and TGF-β1 in rat serum were detected by ELISA.Results:Compared with blank group,the nasal mucosa of the model group showed continuous interruption,structural destruction,cilia shedding,and goblet cell proliferation.Compared with model group,the pathological changes of nasal mucosa in drug intervention groups showed varying degrees of mitigation.Compared with blank group,the contents of IL-10,Foxp3 mRNA in nasal mucosa and IL-10,TGF-β1 in serum were significantly decreased in model group(P<0.05),while the contents of IL-17,RORγt mRNA,RORγt mRNA/Foxp3 mRNA in nasa mucosa and IL-17 in serum were significantly increased in model group(P<0.05).After drug intervention,compared with model group,the levels of IL-10,Foxp3 mRNA in nasal mucosa and IL-10,TGF-β1 in serum were increased in drug intervention groups(P<0.05),while the levels of RORγt mRNA,RORγt mRNA/Foxp3 mRNA in nasal mucosa and IL-17 in serum were decreased in drug intervention groups(P<0.05).Compared with model group,the expression of IL-17 in nasal mucosa was significantly decreased in western medicine group and the high-dose Chinese medicine group(P<0.05).Conclusion:Lu’e Biyan Formula can alleviate the nasal allergic symptoms of rats,regulate the immune state of the body by regulating the balance of Th17/Treg in vivo,and finally achieve the purpose of controlling the inflammatory response.
