止消温肾宁颗粒对糖尿病肾病大鼠的治疗作用及机制研究

摘　　要：目的:探讨止消温肾宁颗粒对糖尿病肾病(DN)模型大鼠的治疗作用及其作用机制。方法:将180只SD大鼠随机分为假手术组(剥除左肾脂肪薄膜),模型组(单侧肾切联合腹腔注射链脲佐菌素),止消温肾宁低、中、高剂量组,渴络欣组与厄贝沙坦组。造模后第4周开始灌胃给药,连续给饲12周。取材后检测尿蛋白/尿肌酐的比值(ACR)、24 h尿蛋白、空腹血糖(FBG)、血清尿素氮(BUN)、糖化血清蛋白(GSP)、低密度脂蛋白胆固醇(LDL-C)、甘油三脂(TG)及晚期糖基化终末产物(AGEs)的含量,肾组织病理状况及肾皮质结缔组织生长因子(CTGF)蛋白表达。结果:与模型组比较,止消温肾宁颗粒可降低ACR、24 h尿蛋白、FBG、BUN、GSP、LDL-C、TC及AGEs,并可抑制CTGF的过高表达,有效缓解肾小球及肾小管病变。结论:止消温肾宁颗粒对DN大鼠有较好的治疗作用,其机制可能与调节血清与尿液生化指标、缓解肾脏组织病变有关。Objective:To explore the therapeutic effect and mechanism of Zhixiao Wenshenning Granules on diabetic nephropathy(DN)model rats.Methods:One hundred and eighty SD rats were randomly divided into the sham operation group(stripping left renal fat membrane),the model group(unilateral nephrectomy combined with intraperitoneal injection of Streptozotocin),the Zhixiao Wenshenning Granules low-,medium-,and high-dose group,the Keluoxin group and the Irbesartan group.The rats were given intragastric administration from the 4th week after the establishment of the model,and were fed for 12weeks continuously.After sampling,the ratio of urinary protein to creatinine(ACR),24 h urinary protein,fasting blood glucose(FBG),serum urea nitrogen(BUN),glycosylated serum protein(GSP),low density lipoprotein cholesterol(LDL-C),triglyceride(TG)and advanced glycation end products(AGEs),the pathological status of tissue and the expression of connective tissue growth factor(CTGF)proteins were detected.Results:Compared with the model group,Zhixiao Wenshenning Granules could reduce ACR,24 h urinary protein,FBG,BUN,GSP,LDL-C,TC and AGEs,and inhibit the overexpression of CTGF in renal cortex.It can effectively relieve the pathological changes of glomeruli and renal tubules.Conclusion:Zhixiao Wenshenning Granules has a good therapeutic effect on DN model rats,and its mechanism may be related to the regulation of serum and urine biochemical indexes and relief of renal pathological changes.
