摘  要:【目的】探究YUCCA家族基因在葡萄穗梗褪绿过程中表达模式。【方法】通过生物信息学的方法,分析了葡萄YUCCA家族基因聚类、内含子/外显子结构、蛋白保守基序和启动子顺式元件,通过液相色谱质谱联用的方法测定阳光玫瑰葡萄穗梗褪绿过程中IAA含量的变化,利用Real-time PCR检测YUCCA家族基因在穗梗褐变过程中的表达趋势。【结果】从葡萄基因中鉴定得到10个YUCCA家族基因,命名为VvYUCCA 1~VvYUCCA10。葡萄YUCCA家族基因可以分为4个亚家族;同一个亚家族的基因含有相似的内含子/外显子结构和蛋白保守基序;在YUCCA家族基因的启动子中,存在大量脱落酸、茉莉酸和水杨酸等信号响应元件;IAA含量在穗梗褪绿过程中明显升高。【结论】YUCCA 1和YUCCA8的表达与IAA含量呈正相关关系,可能参与葡萄采后贮藏过程中穗梗的褪绿。【Objective】To explore expression pattern of YUCCA gene family during rachis degreening of grape.【Methods】Phylogenetic relationship,exon/intron structural and conserved protein domains and promoter subcis element of grape YUCCA gene family were analyzed.IAA content during rachis degreening was quantified by liquid chromatography and mass spectrometry,and the expression pattern of YUCCA gene family during rachis degreening of grape were detected by real-time PCR.【Results】Genome-wide identification of YUCCA gene family in grape was performed,and 10 genes were identified and named from VvYUCCA 1-VvYUCCA10.The result indicated that:YUCCA gene family in grape can be divided into 4 subfamilies;similar exon/intron structures and conserved domains existed in the same subfamily;lots of ABA,MeJA and SA response cis-element were found in promoters of grape YUCCA gene family;the content of IAA was significantly significantly increased during rachis degreening.【Conclusion】VvYUCCA 1 and VvYUCCA 8 showed positive correlation with IAA content,indicating that they might participate in rachis degreening of grape.
