柴胡疏肝散对围绝经期综合征肝郁证大鼠行为学及血清T、PROG含量的影响

摘　　要：目的研究柴胡疏肝散对围绝经期综合征肝郁证模型大鼠行为学的影响及对血清中PROG(孕酮)、T(睾酮)含量的影响。方法选取30只雌性SD大鼠,随机分成正常组、模型组和给药组,采用双侧卵巢去势手术加上经典的CUMS(慢性温和不可预知刺激)方法建立围绝经期综合征肝郁证的大鼠模型,用柴胡疏肝散在造模同时进行灌胃干预,共计21 d。造模前后2次进行行为学实验;实验结束后采用ELISA法(酶联免疫吸附法)测定各组大鼠血清中相应指标PROG(孕酮)、T(睾酮)的含量。结果在旷场实验结果中,模型组的移动总距离和直立次数较正常组均下降(P<0.05),给药组的移动总距离比模型组显著增加(P<0.01),给药组直立次数比模型组增多(P<0.05);在强迫游泳实验中,各组大鼠的游泳不动时间无差异(P>0.05);ELISA实验结果中,模型组的睾酮、孕酮水平比正常组均下降(P<0.01);给药组大鼠血清睾酮含量比模型组显著提高(P<0.01),给药组血清孕酮含量与模型组比较,差异无统计学意义(P>0.05)。结论柴胡疏肝散可改善围绝经期综合征肝郁证大鼠的抑郁样行为,提高大鼠血清中的睾酮激素水平。Objective:To study the effect of Chaihu Shugan Powder on the behavior of rats with perimenopausal syndrome and liver stagnation syndrome and the effect on the level of PROG(progesterone)and T(testosterone)in serum.Methods:30 female SD rats were selected and randomly divided into normal group,model group and administration group.Bilateral ovariectomy combined with classical CUMS(chronic mild and unpredictable stimulation)method was used to establish rat model with perimenopausal syndrome and liver stagnation syndrome.Chaihu Shugan Powder was used for gavage intervention at the same time as the model was established,for 21 days.Behavioral experiments were conducted twice before and after modeling.After the experiment,ELISA(enzyme-linked immunosorbent assay)was used to determine the contents of corresponding indexes PROG(progesterone)and T(testosterone)in the serum of rats in each group.Results:In the open field test results,the total moving distance and standing times of the model group were decreased than those of the normal group(P<0.05).The total moving distance of the administration group was obviously increased than that of model group.The times of standing uprights of the administration group was more than that of the model group(P<0.05).In the forced swimming test,there was no difference in the swimming immobility time of the rats in each group(P>0.05).In the ELISA test results,the levels of testosterone and progesterone in the model group were decreased than those in the normal group(P<0.01).Compared with that of the model group,the levels of serum testosterone in the administration group were significantly increased(P<0.01).There was no statistically significant difference in serum progesterone levels between the administration group and the model group(P>0.05).Conclusion:Chaihu Shugan Powder can improve the depression-like behavior of rats with perimenopausal syndrome and liver stagnation syndrome and increase the level of testosterone in serum of rats.
