摘  要:香菇是一种重要的食用菌,温度是影响香菇菌丝生长的重要因素,每年有大面积的香菇遭受到寒冷温度的威胁,导致其产量下降。OsICE1是水稻中的一种冷调控关键蛋白,在水稻抗寒种质创制中具有重要作用。本研究从日本晴水稻的芽中提取了RNA并克隆得到OsICE1的编码区序列,构建pCsV1300-OsICE1的表达载体,并成功完成香菇菌丝体的遗传转化,获得了2个转基因系。本研究为OsICE1在耐冷真菌种质创制中的潜在作用提供参考,也为香菇菌丝体抗寒冷的分子育种方面奠定基础。Lentinus edodes is an important edible fungus,and temperature is an important factor affecting the mycelial growth of Lentinus edodes,every year,a large area of Lentinus edodes is threatened by cold temperature,which leads to the decline of its yield.OsICE1is a key protein of cold regulation in rice,which plays an important role in the creation of cold resistant germplasm.In this study,RNA was extracted from the buds of Japonica Rice and the coding region of OsICE1 was cloned to construct the expression vector pcsv1300-OsICE1,the genetic transformation of Lentinus edodes mycelium was successfully completed,and two transgenic lines were obtained.This study provides a reference for the potential role of osice1 in cold tolerant fungal germplasm creation,and lays a foundation for molecular breeding of Lentinus edodes mycelium cold tolerance.
