青花菜高温和盐胁迫相关LncRNAs的鉴定与表达特征分析

摘  要:为探讨长链非编码RNA在青花菜高温和盐胁迫中的功能。以青花菜高代自交系‘WN12-95B’为试验材料,通过RNA-seq测序、靶基因注释和qRT-PCR分析,筛选出与青花菜高温和盐胁迫相关的LncRNA。在青花菜高温和盐胁迫文库中鉴定到2432条LncRNAs,筛选获得158个差异LncRNAs,其中96个上调表达,62个下调表达。序列特征分析发现LncRNAs的长度较mRNA短,外显子个数也比mRNA少。靶基因注释显示差异LncRNAs可通过谷胱甘肽代谢、信号转导以及氧化磷酸化等参与青花菜高温和盐胁迫响应。qRT-PCR分析结果显示10个挑选的差异LncRNAs在盐胁迫和高温胁迫中的表达趋势各具特点。XLOC_033957、XLOC_034964、XLOC_005515、XLOC_027154、XLOC_023592和XLOC_035830在高温胁迫下呈现先下降再上升的模式,但在盐胁迫中其表达模式存在差异。XLOC_033671、XLOC_003826、XLOC_06988和XLOC_024730在盐胁迫下呈持续上调表达,在高温胁迫下先上升后下降。通过测序及数据分析,鉴定出青花菜高温和盐胁迫相关的LncRNA的相关基因,为进一步分析LncRNAs在青花菜高温和盐胁迫下的调控机制提供一定的基础。To clarify the regulatory functions of long non coding RNA under high temperature and salt stress in broccoli,the high generation inbred line‘WN12-95B’of broccoli was used as experimental material,then LncRNAs related to high temperature and salt stress in broccoli were selected through RNA-seq sequencing,target gene annotation and qRT-PCR analysis.2432 LncRNAs were identified in high temperature and salt stress library of broccoli.158 different expression LncRNAs were screened out,of which 96 were up-regulated and 62 were down-regulated.Sequence analysis showed that LncRNAs had shorter length and fewer exons than mRNAs.The target genes annotation showed that these LncRNAs could participate in high temperature and salt stress could response through glutathione metabolism,signal transduction,oxidative phosphorylation and other biological processes in broccoli.qRT-PCR analysis showed that the expression features of 10 seleceted LncRNAs exhibited diverse patterns under above stresses.XLOC_033957,XLOC_034964,XLOC_005515,XLOC_027154,XLOC_023592 and XLOC_035830 first decreased and then increased under high temperature stress,but their expression patterns were diverse under salt stress.XLOC_033671,XLOC_003826,XLOC_06988 and XLOC_024730 were continuously up-regulated under salt stress.However,it increased first and then decreased under high temperature stress.The sequencing and data analysis had identified LncRNAs related to high temperature and salt stress in broccoli,which could provide basis for further study on regulatory mechanism of these LncRNAs under high temperature and salt stress in broccoli.
