摘  要:【目的】分离侵染贵州火龙果的蟹爪兰X病毒(Zygocactus virus X,ZyVX)和火龙果X病毒(Pitaya virus X,PiVX)基因组序列,探讨其序列差异和遗传变异。【方法】利用RT-PCR筛选含有ZyVX和PiVX的火龙果样品,然后通过RNA-seq获得ZyVX和PiVX基因组相关序列,进行序列组装、一致性比较、基因重组和系统进化分析。【结果】获得4条ZyVX贵州分离物和3条PiVX贵州分离物基因组近全长序列,长度分别为6570~6600和6649~6657 bp,分别覆盖参考基因组序列的99.2%~99.6%和99.3%~99.8%。4条ZyVX贵州分离物之间的基因组序列一致性为96.7%~99.5%,与其他已报道分离物的序列一致性为91.4%~96.9%,其ORF编码的氨基酸序列一致性为93.8%~100%。3条PiVX贵州分离物之间基因组序列一致性为98.2%~98.5%,与其他已报道分离物序列一致性为98.0%~99.1%,其ORF编码的氨基酸序列一致性为97.8%~100%。ZyVX和PiVX贵州分离物基因重组分析表明未发现明显的重组事件,系统进化分析表明未发生明显的遗传分化。【结论】当前贵州ZyVX和PiVX群体的基因组序列一致性高,其变异均为单核苷酸变异,未发生基因重组和遗传分化。【Objective】The genomic sequences of Zygocactus virus X(ZyVX)and Pitaya virus X(PiVX)infecting pitaya in Guizhou province were isolated,and their sequence differences and genetic variation were investigated.【Method】The pitaya samples containing ZyVX and PiVX were screened by RT-PCR,and then ZyVX and PiVX genome sequences were obtained by RNA-seq,sequence assembly,identity comparison,gene recombination and phylogenetic analysis were performed.【Result】The nearly full-length genome sequences of four ZyVX Guizhou isolates and three PiVX Guizhou isolates were obtained with lengths of 6570-6600 and 6649-6657 bp,respectively,which covered 99.2%-99.6% and 99.3%-99.8%of the reference genome sequence,respectively.The genomic sequence identities among four ZyVX Guizhou isolates were 96.7%-99.5%,and the sequence identities with other reported isolates were 91.4%-96.9%.The amino acid sequence identities encoded by ORF of ZyVX Guizhou isolates were 93.8%-100%.The genomic sequence identities between three PiVX Guizhou isolates were 98.2%-98.5%,and the sequence identities with other reported isolates were 98.0%-99.1%.The amino acid sequence identities encoded by ORF of PiVX Guizhou isolates were 97.8%-100%.Genetic recombination analysis showed that no obvious recombination events were found in the genomic sequence of ZyVX Guizhou isolates and PiVX Guizhou isolates,and phylogenetic analysis suggested that there were no obvious genetic differentiation events.【Conclusion】The genome sequence identities of current ZyVX and PiVX population in Guizhou province were high,whose existing mutations were single nucleotide mutations,and no genetic recombination and genetic differentiation had occurred.
