摘  要:目的 探究化妆品中药原料紫草素和姜黄素在动物替代试验中的应用。方法 选用紫草素和姜黄素单体原料分别进行体外兔角膜上皮细胞短时暴露试验,体外3T3中性红摄取光毒性试验与皮肤变态反应:局部淋巴结试验(DA法,LLNA:DA)。结果 紫草素在3个试验中结果均为阴性。姜黄素不能在兔角膜上皮细胞短时暴露试验和体外3T3中性红摄取光毒性试验规定的溶媒中均匀溶解,只进行了LLNA:DA试验,结果为阴性。结论 动物替代试验结果显示紫草素眼刺激、光毒性和致敏性为阴性,姜黄素致敏性为阴性。姜黄素和紫草素在规定溶媒中的溶解性决定了二者在动物替代试验中的适用性,在进行动物替代试验前需对样品进行充分的溶解度预试验。OBJECTIVE To study the application of shikonin and curcumin in animal substitution experiments as the raw materials of cosmetics.METHODS The raw materials of shikonin and curcumin in cosmetics were separately carried out with shortterm exposure test(STE test) in rabbit corneal epithelial cells,3T3 cell neutral red uptake test and murine local lymph node assay:Daicel adenosine triphosphate(LLNA:DA) test method.RESULTS The results were negative predition in the three tests for shikonin.Curcumin could not be uniformly dissolved in the solvent specified in the short-term exposure test of rabbit corneal epithelial cells and in the 3T3 cell neutral red uptake test.Only the LLNA:DA test was performed and the result was negative predition.CONCLUSION Animal substitution experiment results from eye irritation,skin phototoxicity and sensitization were negative predition for shikonin,and curcumin was negative predition for sensitization.The solubility of curcumin and shikonin in the prescribed solvent determine their applicability in animal substitution experiments.Before conducting animal substitution experiments,sufficient solubility pre-tests should be performed on the samples.
