摘　　要：目的探讨大鼠局灶性脑缺血再灌注(MCAO/R)损伤模型大鼠脑组织miR-211-5p表达情况。方法取SPF级成年大鼠40只,分为假手术组(Sham组)、MCAO/R+溶剂组(Vehicle组)、MCAO/R+miR-211-5p组(Mimic组)、MCAO/R+anti-miR-211-5p组(Antagomir组),每组10只。评价各组大鼠神经功能缺损评分,检测各组大鼠脑梗死体积及miR-211-5p、脑源性神经营养因子(BDNF)蛋白表达水平。结果Vehicle组存活率低于Sham组,Longa评分升高,miR-211-5p处理后存活率升高,Longa评分降低(P<0.05)。Mimic组脑梗死体积百分比较Vehicle组降低,Antagomir组脑梗死体积百分比高于Mimic组(P<0.05)。与Sham组和Vehicle组比较,Mimic组miR-211-5p表达量升高,Antagomir组表达量降低,Antagomir组miR-211-5p表达量低于Mimic组(P<0.05)。Mimic组BDNF表达量显著高于其他3组,Antagomir组BDNF表达量最低(P<0.05)。结论miR-211-5p表达与大鼠MCAO/R损伤相关。Objective To investigate the expression of miR-211-5p in brain tissue of rats with focal cerebral ischemia-reperfusion(MCAO/R)injury model.Methods Forty SPF adult rats were divided into sham operation group(Sham group),MCAO/R+solvent group(Vehicle group),MCAO/R+miR-211-5p group(Mimic group)and MCAO/R+anti-miR-211-5p group(Antagomir group),with 10 rats in each group.The neurological deficit score of rats in each group was evaluated.The cerebral infarction volume of rats in each group,and the expression levels of miR-211-5p and brain-derived neurotrophic factor(BDNF)protein were detected.Results The survival rate of Vehicle group was significantly lower than that of Sham group,the Longa score increased significantly,while the survival rate increased and the Longa score decreased after miR-211-5p treatment(P<0.05).The percentage of cerebral infarction volume was lower in Mimic group than in Vehicle group,and higher in Antagomir group than in Mimic group(P<0.05).Compared with Sham group and Vehicle group,the expression of miR-211-5p increased in Mimic group and decreased in Antagomir group;the expression of miR-211-5p in Antagomir group was lower than that in Mimic group(P<0.05).The expression of BDNF in Mimic group was significantly higher than that in the other three groups,which was the lowest in Antagomir group(P<0.05).Conclusion The expression of miR-211-5p was correlated with MCAO/R injury in rats.
