摘  要:目的:观察小鼠丘脑室旁核(PVT)向边缘系统部分区域的分支投射,并为其参与痒信息的传递提供形态学证据。方法:小鼠分为两组。将荧光金(FG)分别注入第1组动物的内侧前额叶皮质(mPFC)、岛叶(IC)或杏仁中央核(CeA);将FG和霍乱毒素B亚单位(CTB)分别注射到第2组小鼠的两个不同核团(mPFC/IC、mPFC/CeA和IC/CeA);灌注固定前2 h再给予动物急性痒刺激。结合免疫荧光组织化学染色技术观察小鼠PVT内向mPFC、IC或CeA投射、分支投射神经元的分布及其表达FOS蛋白的状况。结果:PVT内存在向mPFC、IC和CeA投射、分支投射并表达FOS的神经元,这些标记神经元在PVT内的分布和比例存在差异。结论:PVT内具有定位分布倾向的部分神经元借助分支投射同时向边缘系统的不同区域传递痒信息。Objective:To study the neuronal collateral projections of paraventricular thalamic nucleus(PVT)to certain regions of the limbic cortex in mice,and provide morphololgical evidence for its participation in itch infornation transmission.Methods:The mice were divided into two groups.For group 1 mice,fluoro-gold(FG)was injected into the medial prefrontal cortex(mPFC),insular cortex(IC)and central amygdaloid nucleus(CeA),respectively.For group 2 mice,FG and cholera toxin subunit B(CTB)were injected into the mPFC and IC,mPFC and CeA,IC and CeA,respectively.Animals were given acute itch stimulation 2 hours before perfusion.Immunofluorescence histochemical staining was used to examine the distributions of the projection,collateral projection neurons and FOS protein expression in neurons induced by acute itch stimulation in PVT.Results:Distinct distributions and percentages of the projection and collateral projection neurons of which FOS were also expressed within them were observed in PVT.Conclusion:Some PVT neurons simultaneously transmitting itch information to certain regions of the limbic system by collateral projection with tendency of topographic distribution pattern.
