摘 要:目的建立同时测定血浆中血管紧张素Ⅰ(AngⅠ)、血管紧张素Ⅱ(AngⅡ)和醛固酮(Aldo)的液相色谱串联质谱(LCMS/MS)法。方法将200μL血浆和温育液在37℃温育3 h后,加入同位素内标,用固相萃取法提取各激素。通过LCMS/MS分析AngⅠ、AngⅡ和Aldo,最终用标准曲线定量。结果AngⅠ、AngⅡ和Aldo的线性范围分别为0.2~50.0、0.002~0.500、0.02~5.00 ng/mL,定量限(LOQ)为0.2、0.002、0.02 ng/mL。3种分析物的回收率在91%~103%之间。方法灵敏度高、重现性好,变异系数(CV)分别为1.4%~4.6%(AngⅠ)、2.1%~5.1%(AngⅡ)、2.4%~5.2%(Aldo)。健康人血浆中AngⅠ、AngⅡ、Aldo的浓度范围分别为1.4~11.8、0.003~0.016、0.02~0.20 ng/mL。方法无明显基质效应和携带污染。结论建立了一种可同时准确、快速测定血浆AngⅠ、AngⅡ和Aldo的LC-MS/MS方法。Objective To establish a liquid chromatography tandem mass spectrometry(LCMS/MS)method for simultaneous determination of angiotensinⅠ(AngⅠ),angiotensinⅡ(AngⅡ)and aldosterone(Aldo)in plasma.Methods After 200μL of plasma and incubation solution were incubated at 37℃for 3 hours,an isotope internal standard(IS)was added into the mixture and each hormone was extracted by solid phase extraction.AngⅠ,AngⅡand Aldo were determined by LCMS/MS,and finally quantified by standard curve.Results The linear ranges of AngⅠ,AngⅡand Aldo were 0.2 to 50.0,0.002 to 0.500 and 0.02 to 5.00 ng/mL,respectively,and the low limits of quantification(LOQ)were 0.2,0.002,0.02 ng/mL,respectively.The recovery rates of the three analytes were between 91%and 103%.The developed method presented high sensitivity and good reproducibility.The coefficient of variation(CV)were 1.4%to 4.6%(AngⅠ),2.1%to 5.1%(AngⅡ)and 2.4%to 5.2%(Aldo).The concentration ranges of AngⅠ,AngⅡand Aldo in normal human plasma were 1.4 to 11.8,0.003 to 0.016 and 0.02 to 0.20 ng/mL,respectively.No obvious matrix effect and carryover pollution was found.Conclusion An accurate and rapid LCMS/MS method for simultaneous determination of AngⅠ,AngⅡand Aldo in plasma was established.
