摘 要:目的考察冷藏加压对于腊叶标本成品保绿的影响。方法随机选择6种植物在增大压力和降低温度的8组条件下进行腊叶标本的压制,压制成功后,通过DMSO-80%丙酮两步快速浸提法及紫外分光光度法,对标本植物叶的叶绿素含量进行测定,计算出叶绿素含量,采用多因素方差分析,对结果进行比较,考察温度和压力对于标本的保绿是否有显著影响。结果温度和压力的主效应,及温度与压力两因素的交互效应均对叶绿素的含量有显著影响(P<0.05)。结论冷藏2-8℃加压相较于自然干燥法对于腊叶标本的保绿效果更好,但根据植物品种的不同,压力值各不相同。OBJECTIVE To investigate the effects of reducing temperature and increasing pressure on the green preservation of wax leaf specimens.METHODS Six kinds of plants were selected randomly and pressed under 8 conditions of increasing pressure and decreasing temperature.After pressing successfully,the chlorophyll content of the finished leaves was determined by DMSO-80% acetone two-step rapid extraction method and UV spectrophotometry,and the chlorophyll content was calculated.The results were compared by multi factor analysis of variance,and the temperature and pressure were investigated Whether the force has a significant effect on the green preservation of specimens.RESULTS The main effects of temperature and pressure,and the interaction of temperature and pressure had significant effects on the content of chlorophyll(P<0.05).CONCLUSION compared with natural drying method,2-8℃ cold storage pressure method has better green preservation effect on wax leaf specimens,but the pressure values are different according to different plant varieties.
