摘 要:目的分析福建不同种源枇杷叶的遗传多态性。方法CTAB改进法提取枇杷叶总基因组DNA,采用ISSR分子标记分析遗传多态性,Arlqeuln和NTSYS-pc version 2.10e软件计算遗传距离并进行聚类分析。结果从100对ISSR引物中筛选出14对多态性丰富的引物,共扩增出79条多态性条带,占扩增总带数的52.67%,每个多态性引物多态性比例41.67%~61.53%。24份枇杷叶样本的遗传相似系数在0.1052~0.9473之间,分可为五个分支,遗传分化明显。枇杷叶的亲缘关系与地理分布的相关性不显著。结论不同种源枇杷叶具有显著的遗传多样性,ISSR可作为构建枇杷叶DNA指纹图谱的有效工具之一。OBJECTIVE To investigate the genetic diversity of Folium eriobotryae from different strains in Fujian Province.METHODS Genomic DNA of Folium eriobotryae were extracted by CATB method,and ISSR Molecular marker were applied to analysis of the genetic diversity.The genetic similarity(GS)and cluster analysis were performed by Arlqeuln and NTSYS-pc-version 2.10 e softwar,respectively.RESULTS Of the 100 primers screened,14 primers produced highly reproducible ISSR bands,using these primers,79 discernible DNA fragments were generated with percentage of polymorphic ranging from 41.67% to 61.53%.According to GS,which ranged from 0.1052 to 0.9473,samples of Folium eriobotryae from different strains can be divided into five groups.The relationship between genetic distance and geography was not significant.CONCLUSION There were considerable genetic variation among Folium eriobotryae from different strains in Fujian Province.
