灵芝总黄酮提取物的制备及其细胞毒活性

摘　　要：目的从灵芝中制备总黄酮,并探讨其体外细胞毒活性。方法回流提取法结合聚酰胺柱色谱法从灵芝中制备总黄酮提取物;紫外-可见分光光度法测定醇浸膏及总黄酮提取物中总黄酮含量;MTT法测试灵芝总黄酮提取物的细胞毒活性。结果灵芝醇提浸膏与聚酰胺按质量比为1∶1混匀后装柱,先用40%乙醇洗脱,再用无水乙醇洗脱,收集无水乙醇洗脱液,减压浓缩蒸干得灵芝总黄酮提取物。醇浸膏中含总黄酮含量为8.98%,灵芝总黄酮提取物中总黄酮含量为18.09%。灵芝总黄酮提取物在体外对人胰腺癌MIA-PACA-2和人乳腺癌MDA-MB-231肿瘤细胞的增殖有抑制作用,IC_(50)分别为(123.7±4.1)μg·mL^(-1)和(196.1±2.5)μg·mL^(-1)。对HepG2、SMM C7721和HL-60细胞的增殖抑制作用较弱。结论聚酰胺可富集灵芝中总黄酮,灵芝总黄酮提取物对人胰腺癌MIA-PACA-2细胞和人乳腺癌MDA-MB-231细胞有一定细胞毒活性。OBJECTIVE To investigate in vitro cytotoxic activity of the total flavonoids extract prepared from Ganoderma lucidum.METHODS Total flavonoids were extracted from Ganoderma lucidum by reflux extraction combined with polyamide column chromatography.The content of total flavonoids in alcohol extract and total flavonoids extract was determined by UV-vis spectrophotometry.MTT assay was used to determine the cytotoxic activity of total flavonoids extract from Ganoderma lucidum.RESULTS Ganoderma lucidum alcohol extract and polyamide were mixed at mass ratio of 1∶1 and then loaded into a chromatography column.The polyamide column was eluted with 40% ethanol,and then eluted with anhydrous ethanol.The total flavonoid extract of Ganoderma lucidum was prepared by collecting anhydrous ethanol eluent under reduced pressure.The content of total flavonoids in alcohol extract and total flavonoids extract was 8.98% and 18.09%,respectively.Ganoderma lucidum total flavonoids extract inhibited the proliferation of MIA-PACA-2 and MDA-MB-231 tumor cells in vitro,with IC_(50) values of(123.7±4.1)μg·mL^(-1) and 196.1±2.5μg·mL^(-1),respectively.The inhibitory effect on the proliferation of HepG2,SMMC7721 and HL-60 was weak.CONCLUSION Polyamide can enrich total flavonoids in Ganoderma lucidum.Ganoderma lucidum flavonoids extract showed certain cytotoxic activity against human pancreatic cancer MIA-PACA-2 cells and human breast cancer MDA-MB-231 cells.
