摘 要:目的:探讨荔仁总皂苷体内抗鸭乙型肝炎病毒(DHBV)感染及其护肝作用。方法:在建立鸭乙肝动物模型后,选取DHBV感染阳性幼鸭60只,按随机数字表法分为5组:模型对照组(喂蒸馏水)、阿昔洛韦阳性对照组(ACV组)和荔仁总皂苷高、中、低剂量组(每组12只),每日灌胃给药1次,连续给药14 d,同时分别在给药前、给药后7 d、14 d及停药后5 d,抽取鸭子血液,检测每组鸭子血清样本中的鸭HBV DNA(DHBV-DNA)拷贝量、鸭乙肝表面抗原(DHBsAg)和鸭乙肝病毒e抗原(DHBeAg)的OD值及天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)活性的变化。实验结束时取肝脏组织苏木精-伊红(HE)染色后观察其病理学改变。结果:荔仁总皂苷高、中、低剂量组均可有效抑制DHBV-DNA的复制并呈量效关系(P<0.01);同时荔仁总皂苷高剂量组能够抑制停药后的病毒反跳,其抑制效果优于ACV组。另外,荔仁总皂苷高剂量组还能降低AST活性(P<0.05)。肝脏病理检查显示,与模型对照组比较,荔仁总皂苷高、中剂量组能减轻鸭子感染病毒后肝脏内的炎性病变,有效改善肝组织病理损害。结论:荔仁总皂苷具有一定的抗鸭乙肝病毒、降酶以及改善肝脏病理的作用。Objective:To explore the duck hepatitis B virus(DHBV)infection and its hepatocyte protection effects of total saponins of Litchi Semen.Methods:After the establishment of DHBV animal model,60 DHBV positive baby ducks were randomly divided into 5 groups(n=12 in each group):model control group(fed with distilled water),acyclovir positive control group(ACV group)and high,middle and low dose groups of total saponins of Litchi Semen.The ducks were given intragastric administration once a day for 14 consecutive days.At the same time,ducks’blood samples were taken before administration,7 days,14 days and 5 days after drug withdrawal.The copy amount of DHBV-DNA,the OD value of DHB surface antigen(DHBsAg)and DHBV e antigen(DHBeAg)and the activity changes of aspartate aminotransferase(AST)and alanine aminotransferase(ALT)in duck serum samples were measured.At the end of the experiment,the liver tissue was stained with hematoxylin-eosin(HE)to observe the pathological changes.Results:The high,middle and low dose groups of total saponins of Litchi Semen could effectively inhibit the replication of DHBV-DNA in a dose-dependent manner(P<0.01).The high dose group could inhibit the rebound of virus after drug withdrawal,and the inhibitory effect was better than that of ACV group.In addition,the high dose group could also reduce the activity of AST(P<0.05).Liver pathological examination showed that,compared with the model group,the high and middle dose groups of total saponins of Litchi Semen could reduce the inflammatory lesions in the liver of ducks infected with virus and effectively improve the pathological damage of liver tissue.Conclusion:The total saponins of Litchi Semen have a certain effect on anti-duck hepatitis B virus,lowering enzymes and improving liver pathology.
