苏木乙酸乙酯提取物对肺动脉高压模型大鼠血清TXA2、PGI2及肺组织病理形态学的影响

摘　　要：目的观察苏木乙酸乙酯提取物对肺动脉高压模型大鼠血清TXA2、PGI2及肺组织病理形态学的影响,探讨其对PAH保护作用及机制研究。方法将50只雄性SD大鼠随机分为空白组、模型组、高剂量组、低剂量组、对照组5组,模型制备采用腹腔注射MCT;高剂量组、低剂量组分别给予苏木乙酸乙酯提取物灌胃;对照组给予阿魏酸钠灌胃。实验期满测平均肺动脉压(mPAP),测定血清TXA2与PGI2水平,用光镜观察肺组织及肺小动脉病理形态学变化。结果与模型组相比,高剂量组和低剂量组大鼠肺组织炎性渗出减轻,肺小动脉壁增生缓解,管腔变窄好转;高剂量组、低剂量组和对照组mPAP及血清TXA2水平显著降低,血清PGI2水平显著升高,差异有统计学意义(P<0.05)。结论苏木乙酸乙酯提取物能减轻PAH模型大鼠肺组织及肺小动脉的病理改变,降低PAH模型大鼠血清TXA2水平,升高PGI2水平,改善血管内皮功能。Objective To observe the effects of ethyl acetate extract of sappan wood on serum thromboxane A2(TXA2),prostacyclin(PGI2)and lung histopathology in rats with pulmonary arterial hypertension(PAH),and to explore the protective effect and mechanism of ethyl acetate extract of sappan wood on PAH.Methods 50 male SD rats were randomly divided into 5 groups:blank group,model group,high-dose group,low-dose group and control group.The model was established by intraperitoneal injection of MCT.The high-dose group and the low-dose group were given ethyl acetate extract of sappan respectively.The control group was given sodium ferulate by gavage.At the end of the experiment,the mean pulmonary arterial pressure(mPAP)and the levels of TXA2 and PGI2 in serum were measured.The pathological changes of lung tissue and pulmonary arterioles were observed by light microscope.Results Compared with the model group,the inflammatory exudation of lung tissue in high-dose group and low-dose group was reduced,the proliferation of pulmonary arteriole wall was alleviated,and the lumen narrowing was improved.The levels of MPAP and serum TXA2 in high-dose group,low-dose group and control group were significantly lower,and the levels of serum PGI2 were significantly higher(P<0.05).Conclusion The ethyl acetate extract of sappan wood can reduce the pathological changes of lung tissue and pulmonary arterioles in PAH model rats,reduce the serum TXA2 level,increase the serum PGI2 level,and improve the vascular endothelial function.
