摘　　要：目的:建立复方天麻首乌片中2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量的HPLC测定法。方法:选择Agilent 5 TC-C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(19∶81),流速为1.0 mL/min,波长320 nm,柱温30℃,进样量5μL。结果:2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的进样浓度在10.64~340.48 mg/L(r=0.99999)呈良好线性关系;2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的平均回收率(n=6)为95.15%(RSD=2.42%);5个批次复方天麻首乌片样品中2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量测定结果为366.7~856.7μg/g。结论:该法可用于复方天麻首乌片中2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量测定。Objective:To establish a HPLC method for the contents determination of 2,3,5,4'-tetrahydroxystilbene-2-O-β-Dglucoside in fufang tianma shouwu tablets.Methods:The HPLC analysis was performed on a Agilent 5 TC-C18(250 mm×4.6 mm,5μm)column.The mobile phase consisted of acetonitrile and water(19∶81)at the flow rate of 1.0 mL/min.The detection wavelength was 320 nm and the column temperature was 30℃.The sample quantity was 10μL.Results:The contents of 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside had good linear relationship in the ranges of 10.64-340.48 mg/L(r=0.99999).The average recoveries(n=6)were 95.15%with RSD of 2.42%.The contents in 5 sapmles of fufang tianma shouwu tablets were 366.7-856.7μg/g for 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside.Conclusion:According to the methodology validation,this method can be applied to the contents determination of 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside in compound tianma shouwu tablets.
