摘  要:目的运用分子生物技术探究血府逐瘀汤对慢性阻塞性肺疾病(COPD)模型小鼠炎症因子与TLR4/衔接蛋白髓样分化因子(MyD88)/核转录因子-κB(NF-κB)信号通路的作用。方法香烟烟熏联合LPS滴鼻法制作COPD模型小鼠,血府逐瘀汤给药处理后,通过HE染色法镜下比较各组小鼠肺组织病理改变;ELISA检测小鼠血清中炎症因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、白细胞介素-17(IL-17)水平;免疫组化检测小鼠肺组织TLR4、MyD88、NF-κB蛋白水平。结果镜下HE染色可见血府逐瘀汤不同剂量组小鼠小气道、肺泡、血管的病理改变及炎性细胞的聚集均有不同程度地减轻。血府逐瘀汤各剂量组小鼠血清中炎症因子TNF-α、IL-1β、IL-6、IL-8、IL-17的含量较模型组明显下降(P<0.05)。免疫组化观察到血府逐瘀汤各剂量组小鼠肺组织中TLR4、MyD88、NF-κB蛋白表达较模型组更低(P<0.05)。结论血府逐瘀汤可以减少COPD小鼠血清中炎症因子TNF-α、IL-1β、IL-8、IL-17的水平,其机制可能是通过调控TLR4/MyD88/NF-κB信号通路,实现对COPD的治疗作用。Objective:To explore the effects of different concentrations of Xuefu Zhuyu Decoction on inflammatory factors and TLR4/MyD88/NF-κB signaling pathway by molecular biological technology in COPD model mice.Methods:COPD model mice were created by cigarette fumigation combined with LPS nasal drops.After administration of Xuefu Zhuyu Decoction,pathological changes of lung tissues in each group were compared under the microscope by HE staining.The levels of inflammatory cytokines TNF-α,IL-1β,IL-6,IL-8 and IL-17 in serum of mice were detected by ELISA.The protein levels of TLR4,MyD88 and NF-κB in lung tissues of mice were detected by immunohistochemistry.Results:The pathological changes of small airways,alveoli and blood vessels and the aggregation of inflammatory cells in different concentrations of Xuefu Zhuyu Decoction were reduced at HE staining under microscope.Compared with the model group,the contents of TNF-α,IL-1β,IL-6,IL-8 and IL-17in serum of Xuefu Zhuyu Decoction dosage groups were significantly decreased with statistical significance(P<0.05).The expression of TLR4,MyD88 and NF-κB protein in lung tissue of mice in Xuefu Zhuyu Decoction groups were lower than that in the model group by immunohistochemistry(P<0.05).Conclusion:Xuefu Zhuyu Decoction can reduce the levels of inflammatory cytokines TNF-α,IL-1β,IL-8 and IL-17 in the serum of COPD mice,and the mechanism may be through the regulation of TLR4/MyD88/NF-κB signaling pathway,which can realize the therapeutic effect on COPD.
