摘  要:目的探讨柴胡皂苷B2对脂多糖(LPS)诱导的脓毒症肺损伤氧化应激和炎性反应的影响。方法将50只SD大鼠分为空白对照组、模型组和柴胡皂苷B2低、中、高剂量组。空白对照组大鼠腹腔内注射等体积生理盐水;模型组大鼠腹腔注射10 mg/kg的LPS;柴胡皂苷B2低剂量组、中剂量组、高剂量组大鼠腹腔注射10 mg/kg的LPS后,分别注射5、10、20 mg/kg的柴胡皂苷。末次给药24 h后,麻醉处死大鼠,检测肺组织湿干比;TUNEL染色检测肺组织细胞凋亡;Western blotting检测肺组织B淋巴细胞瘤(Bcl-2)、Bcl-2同源二聚体X蛋白(Bax)、p53蛋白表达;酶联免疫吸附测定(ELISA)检测血清内超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-Px)活性、丙二醛(MDA)含量、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)含量。结果与空白对照组相比,模型组内肺组织湿干比、细胞凋亡率、Bax、p53蛋白表达、MDA含量、TNF-α、IL1β、IL-6含量明显升高,Bcl-2蛋白表达、SOD活性、GSH-Px活性明显降低。与模型组相比,低剂量组、中剂量组、高剂量组内肺组织湿干比、细胞凋亡率、Bax、p53蛋白、MDA、TNF-α、IL-1β、IL-6明显降低,Bcl-2蛋白、SOD活性、GSH-Px活性明显升高。结论柴胡皂苷B2能减轻LPS诱导的脓毒症急性肺损伤大鼠氧化应激和炎性反应,并减少细胞的凋亡。Objective:To investigate the effect of saikosaponin B2 on oxidative stress and inflammatory re⁃sponse in septic lung injury induced by lipopolysaccharide(LPS).Methods:A total of 50 SD rats were divided in⁃to five groups,i.e.,the control group,the model group,the low-dose group,the middle-dose group,and the highdose group.The control group was intraperitoneally injected with an equal volume of normal saline;the model group was injected intraperitoneally with 10 mg/kg of LPS;after intraperitoneal injection of 10 mg/kg of LPS in the low-dose group,middle-dose group and high-dose group,saikosaponin B2 were injected with low,medium and high doses of 5,10,and 20 mg/kg.At 24 hours after the last administration,the rats were anesthetized and the lung tissue wet-to-dry ratio was detected;TUNEL staining was used to detect lung tissue cell apoptosis;Western blotting were used to detect lung tissue Bcl-2,Bax,and p53 protein expression;ELISA were used to detect serum SOD activity and GSH-Px activity,MDA content,TNF-α,IL-1β,IL-6 content.Results:Compared with the con⁃trol group,the lung tissue wet-to-dry ratio,apoptosis rate,Bax,p53 protein expression,MDA content,TNF-α,IL1β,IL-6 content were significantly increased in the model group,the levels of Bcl-2 protein expression,SOD activ⁃ity,and GSH-Px activity were significantly reduced.Compared with the model group,lung tissue wet-to-dry ratio,apoptosis rate,Bax,p53 protein expression,MDA content,TNF-αin the low-dose,middle-dose,and high-dose groups,IL-1β,IL-6 content were significantly reduced,the levels of Bcl-2 protein expression,SOD activity,and GSH-Px activity were significantly increased.Conclusion:Saikosaponin B2 can alleviate the oxidative stress and inflammatory response of LPS-induced acute lung injury rats with sepsis and reduce cell apoptosis.
