摘 要:目的:探讨吡咯喹啉醌(PQQ)对急性染镉小鼠肝功能和脂质过氧化的影响。方法:选择48只健康、体重相近的4周龄清洁级小鼠,随机分为4组,每组12只。对照组和染镉模型组均先腹腔注射生理盐水,低、高剂量PQQ干预组分别腹腔注射40、80mg/kgPQQ溶液。2h后,对照组皮下注射生理盐水,其余3组皮下注射35μmol/kg的CdCl2溶液。镉染毒24h后采集血样和肝组织样,用于血清中ALT、AST活力以及肝组织中MDA、GSH含量和SOD、GPx活力的测定。结果:急性染镉能使小鼠血清ALT、AST活性以及肝脏MDA含量与GPx活性显著提高,低、高剂量PQQ干预可以显著降低小鼠血清ALT、AST活性及肝脏MDA含量和GPx活性,而对肝脏SOD活性和GSH含量则无显著的影响。结论:PQQ对急性染镉小鼠肝脏抗氧化系统和肝脏功能具有改善效果,可减轻镉造成的急性肝损伤。Objective:To investigate the effects of pyrroloquinoline quinone(PQQ)on liver function and lipid peroxi⁃dation in acute cadmium-exposed mice.Method:A total of 484-week-old mice with similar body weight were ran⁃domly divided into 4 groups with 12 mice in each.The control group and cadmium-exposed model group were intra⁃peritoneally injected with saline,and the low and high dose PQQ intervention groups were intraperitoneally injected with 40 and 80 mg/kg PQQ solution,respectively.After 2 hours,the control group was subcutaneously injected with saline,and the other 3 groups were subcutaneously injected with 35μmol/kg CdCl2 solution.After 24 h of cadmium exposure,blood samples and liver tissue samples were collected for the determination of ALT and AST activities in serum,MDA and GSH contents and SOD and GPx activities in liver tissue.Result:acute cadmium exposure signifi⁃cantly increased the activities of ALT and AST in serum,MDA content and GPx activity in liver of mice.Low and high dose PQQ intervention significantly decreased the activities of ALT and AST in serum and the content of MDA and GPx in liver of mice,but had no significant effect on the activities of SOD and GSH in liver.Conclusion:PQQ can improve the antioxidant system and liver function of acute cadmium-exposed mice,and alleviate the acute liver injury caused by cadmium.
