摘 要:目的:建立鸡眼草总黄酮大孔树脂纯化工艺。方法:采用紫外分光光度法测定总黄酮含量;采用静态吸附、解吸附实验对6种大孔树脂进行筛选,确定最佳树脂;依据吸附和解吸附条件进行单因素实验,对上样液质量浓度体积、pH、洗脱溶剂体积分数、洗脱液用量、洗脱速度进行考察,确定最佳工艺条件。结果:最佳工艺条件为AB-8型大孔树脂,上样液中总黄酮含量为0.813mg/ml,上样体积为2BV,pH为5,吸附平衡后采用70%乙醇洗脱,洗脱液用量为2.5BV,流速为2BV/h。在此工艺条件下,总黄酮含量可达77.26%,转移率为88.26%,出膏率为7.57%。结论:该工艺稳定可行、简便高效,可用于纯化鸡眼草中的总黄酮。Objective:To explore the purification process of the total flavonoids from Kummerowia striata by macroporous resin.Methods:UV spectrophotometry was used to determine the content of total flavonoids.The adsorption and desorption experiments were optimized for 6 kinds of macroporous resins,and the best resins were selected.According to the adsorption and desorption conditions,single factor experiments were carried out to investigate the mass concentration,volume and pH of loading liquid,volume fraction of elution solvent,dosage of elution solution and elution speed to determine the optimal technological conditions.Result:AB-8 resin was selected as the best resin,and the total flavonoid content in sample solution is 0.813 mg/ml.The loading volume was 2 BV and pH was 5.After adsorption equilibrium,70%ethanol was used for elution with the eluent dosage of 2.5 BV and flow rate of 2 BV/h.Under these conditions,the total flavonoid content reached 77.26%,the transfer rate was 88.26%,and the paste yield was 7.57%.Conclusion:This process is stable,feasible,simple and efficient,and can be used to purify the total flavonoids in Kummerowia striata.
