摘 要:天然抗性相关巨噬蛋白(natural resistance associated macrophage protein,Nramp)在调控植物吸收转运Fe^(2+)、Mn^(2+)、Zn^(2+)和Cd^(2+)等重金属离子方面具有重要作用,其成员OsNramp5基因在锰、铁及镉的吸收和转运过程中扮演重要角色。本研究根据水稻OsNramp5(AB690551.1),同源克隆了富铁锌低镉水稻‘宜香优2115’OsNramp5基因的cDNA全长序列,命名为OsNramp5-YXY2115,并对其序列进行生物信息学分析。研究结果表明,其开放阅读框为1617 bp,编码538个氨基酸,蛋白序列含有12个跨膜结构,分子量约为58.5 kD,理论等电点为6.76。功能结构域分析结果证实,Os Nramp5-YXY2115蛋白包含有Nramp金属离子转运结构域MntH,属于Nramp超家族成员。多序列比对及进化树分析结果表明该序列与NCBI中公布的粳稻Os Nramp5(XP_015645014)氨基酸序列相似性最高,为100%,推测OsNramp5-YXY2115基因可能参与响应Fe^(2+)、Mn^(2+)、Zn^(2+)和Cd^(2+)等重金属胁迫。本研究结果为进一步研究OsNramp5-YXY2115的生物学功能提供了基础资料,也为植物重金属研究提供了重要候选基因。Natural resistance associated macrophage protein(Nramp)plays an important role in regulating the absorption and transportation of heavy metal ions such as Fe^(2+),Mn^(2+),Zn^(2+)and Cd^(2+) in plants.One of its members,OsNramp5plays an important role in the absorption and transportation of manganese,iron and cadmium.Based on the OsNramp5(AB690551.1)fragment sequence,the full-length cDNA sequence of OsNramp5 gene from’Yixiangyou2115’rich in iron,zinc and low cadmium was cloned,named OsNramp5-YXY2115,and its bioinformatics analysis was conducted.The results showed that the c DNA of OsNramp5-YXY2115 contained a 1617 bp ORF encoding a protein of 538 amino acids and it had 12 membrane-spanning domains,with a predicted molecular weight of 58.5 kD and theoretical isoelectric point of 6.76.Functional domain analysis confirmed that the Os Nramp5-YXY2115protein contained the Nramp metal ion transport domain MntH,which belongs to the Nramp superfamily.Multiple sequence alignment and phylogenetic analysis showed that the Os Nramp5-YXY2115 protein shared high similarity(100%)with Os Nramp5(XP_015645014)protein from Oryza sativa Japonica Group,and it is speculated that OsNramp5-YXY2115 may be involved in the response to heavy metal stress such as Fe^(2+),Mn^(2+),Zn^(2+) and Cd^(2+).This study provides basic data for further studying the biological function of OsNramp5-YXY2115,and provide significant candidate genes that respond to the stress of heavy metals in plant.
