摘 要:蛋白磷酸酶2C(PP2C)是植物体内激素信号转导过程中的关键调控酶,在植物逆境胁迫响应和发育过程中发挥重要的作用。本研究以角倍(Rhus chinensis)为试材,通过RT-PCR方法克隆获得RcPP2C基因,其c DNA全长为1013 bp,开放阅读框为330 bp,编码109个氨基酸。RcPP2C蛋白相对分子质量为11.961 kD,不含信号肽和跨膜区,为亲水性非分泌蛋白,主要定位在细胞核。该蛋白的二级结构中主要以α-螺旋和无规则卷曲为主。系统进化分析表明,角倍与川桑(Morus notabilis)、甜橙(Citrus sinensis)亲缘关系最近。基因表达分析显示,RcPP2C在角倍和对照叶片中表达量最高,在有倍叶中表达量最低。从角倍发育的初期至末期,RcPP2C的表达量呈先降低后升高最后又降低的变化规律。RcPP2C基因表达量与ABA含量、角倍体积的相关性分析显示,RcPP2C表达量与ABA含量呈显著负相关,而与角倍体积呈正相关,说明该基因可能参与ABA信号传导途径,通过负调控ABA信号来影响角倍发育。本研究为进一步分析RcPP2C基因在ABA信号传导中的功能以及揭示角倍发育的分子机制提供了基础。Protein phosphatase 2C(PP2C)is a key regulatory enzyme involved in hormone signal transduction in plants,and plays an important role in plant stress response and development.RcPP2C gene was cloned from Rhus chinensis by RT-PCR.The full-length cDNA was 1013 bp,with an open reading frame of 330 bp,encoding 109amino acids.The relative molecular weight of RcPP2C was 11.961 kD,without signal peptide and transmembrane region.It was a hydrophilic non-secretory protein mainly located in the nucleus.The RcPP2C was mainly composed ofα-helix and random coil in secondary structure analysis.Phylogenetic analysis indicated that R.chinensis was the most closely related to Morus notabilis and Citrus sinensis.Gene expression analysis showed that the expression level of RcPP2C was highest in gall and control leaf,and lowest in galled leaf.From the initial to terminal stage of gall development,the expression of RcPP2C showed a change rule of first decreasing,then increasing and finally decreasing.Correlation analysis of expression of RcPP2C with ABA content and gall volume showed that expression of RcPP2C was negatively correlated with ABA content,but positively correlated with gall volume,suggesting that RcPP2C gene may be involved in ABA signaling pathway and negatively regulate ABA signal to affect gall development.This study provides the basis for further analysis of the function of RcPP2C gene in ABA signaling and the molecular mechanism of gall development.
