摘　　要：目的:寻找一种适合黑素细胞的冻存方法,并探讨冻存对其细胞活性的影响。方法:采用3种不同冻存配方冻存黑素细胞,复苏后通过台盼蓝染色检测其细胞存活率、CCK-8检测其细胞活力,并比较不同实验组黑素细胞的酪氨酸酶活性及黑素含量。结果:实验组B[8%二甲基亚砜(DMSO)+92%胎牛血清(FBS)]冻存复苏的黑素细胞存活率[(58.36±6.60)%]及细胞活力[(97.69±2.49)%]均高于其他2组,且复苏后的黑素细胞酪氨酸酶活性及黑素含量与正常传代的黑素细胞比较差异均无统计学意义(P>0.05)。结论:使用8%DMSO+92%FBS冻存复苏黑素细胞后其细胞活性和功能均较好,适宜进行白癜风相关的研究。Objective:To find an appropriate method to freeze cells and study the effect of various methods of cryopreservation on the activity of melanocytes Methods:The melanocytes were frozen by three different methods After cell recovery,the cell viability was tested by trypan blue,and the cytotoxicity by CCK-8.Furthermore,the tyrosinase activity and content of melanin in different treatment groups were compared.Results:The cell viability(5836±6.60)%and cytoactivity(97.69±2.49)%in group B(8%DMSO with 92%FBS)were higher than those in other two groups.There was no significant difference in tyrosinase activity and content of melanin between recovered cells and normal passage cells(P>0.05).Conclusion:The cytoactivity and function are well preserved if the cells are frozen by 8%DMSO with 92%FBS.The frozen melanocytes can be used in vitiligo study.
