薰衣草叶绿体RNA编辑位点的预测与分析

摘  要:RNA编辑是转录后水平的一种调控方式,在植物生长发育、形态建成以及逆境响应等过程具有重要作用。为明确薰衣草(Lavandula angustifolia)叶绿体RNA编辑的组成及其特点,本研究通过生物信息学和PCR,对薰衣草植物叶绿体的RNA编辑位点进行了预测和初步验证。结果发现,薰衣草叶绿体基因组中共预测到44个编辑位点,分布于19个基因上,且所有位点均为C到U的转变。利用PCR确认了ndhB-737是ndhB的基因编辑位点之一。比较薰衣草与罗勒(Ocimum basilicum)、薄荷(Mentha haplocalyx Briq.)、紫苏(Perilla frutescens(L.)Britt.)、迷迭香(Rosmarinus officinalis)、益母草(Leonurus japonicus Houtt)这五种唇形科植物叶绿体RNA编辑位点,发现它们之间存在编辑位点的差异,其中atpF-31、rps2-83、rpoB-113、rpoB-189、rpoB-809、rps14-27、rp120-103、ccsA-187和ndh G-105这9个RNA编辑位点是这六种唇形科植物叶绿体所共有。本研究为薰衣草叶绿体RNA编辑的生物学功能研究奠定基础,为从RNA编辑角度阐明唇形科植物的起源进化关系提供一定依据。RNA editing is a regulation mode at the post-transcriptional level,which plays an important role in plant growth,development,morphogenesis and stress response.In order to clarify the composition and characteristics of chloroplast RNA editing in Lavandula angustifolia,we predicted and initially verified the RNA editing sites in lavender chloroplasts by bioinformatics and PCR.The results showed that a total of 44 editing sites were predicted in lavender chloroplast genome,distributed on 19 genes,and all sites were C to U transition.ndhB-737 was confirmed as one of the gene editing sites of ndhB by PCR.Comparison of the chloroplast RNA editing sites between lavender and five species of labiaceae:Ocimum basilicum,Mentha haplocalyx Briq.,Perilla frutescens(L.)Britt.,Rosmarinus officinalis,and Leonurus japonicus Houtt showed that there were differences in the editing sites of chloroplast RNA.And the nine editing sites of atpF-31,rps2-83,rpoB-113,rpoB-189,rpoB-809,rps14-27,rp120-103,ccsA-187 and ndhG-105 were shared by the chloroplasts of the six labiaceae plants.This study lays a foundation for the study of the biological function of RNA editing in lavender chloroplasts,and provides some basis for clarifying the origin and evolutionary relationship of Labiaceae plants from the perspective of RNA editing.
