摘  要:目的探讨不同年龄大鼠皮肤自然老化中的形态学变化规律及其机制。方法取青年、中年、老年大鼠背部正中皮肤,采用形态学、分子生物学、体视学、电镜等技术观察增龄性皮肤组织中胶原纤维和弹力纤维,胶原原纤维的退化以及基质金属蛋白酶MMPs的相关变化。结果随着年龄增长,皮肤含水量下降(P均<0.05),皮肤胶原蛋白、羟脯氨酸(HYP)含量呈年龄依赖性下降(P均<0.05)。MMP1、MMP12的蛋白和mRNA表达水平随年龄增长递增(P均<0.05)。时序性老化大鼠,表皮最薄,真皮萎缩明显,胶原纤维和弹力纤维含量最低(P均<0.05);超微结构观察,胶原原纤维直径小、间距小、横纹周期短(P均<0.05),密度最大(P<0.05)。结论表皮屏障受损,锁水能力下降,真皮基质金属蛋白酶MMPs激活,胶原纤维和弹力纤维退化,与时序性老化密切相关。Objective To detect the morphology change and underlying mechanism of skin aging in different ages of rats.Methods The dorsal median skin from young,middle-aged and old rats were taken,and the collagenous fiber degeneration and elastic fibrils and the metalloproteinase MMPs in aging skin were observed by morphology,molecular biology,stereology and electron microscope methods.Results With the increase of age,skin moisture content decreased(P all<0.05).The contents of collagen and HYP in skin decreased in an age-dependent manner(P all<0.05).Protein and mRNA expression levels of MMP1 and MMP12 increased with age(P all<0.05).The aged rats had the thinnest epidermis,obvious dermal atrophy,and the lowest content of collagen fiber and elastic fiber(P all<0.05).Ultrastructural observation showed that the diameter and spacing of collagen fibrils were small,and the transverse cycle of collagen fibrils was short(P all<0.05),the density of collagen fibrils was maximam(P<0.05).Conclusion The epidermal barrier was damaged,the water-holding capacity was decreased,the dermal matrix metalloproteinases(MMPs)were activated,and the degradation of collagen fibers and elastic fibers was closely related to sequential aging.
