摘  要:试验旨在确定辽宁西部地区绵羊住肉孢子虫的虫体种类和遗传进化关系,采集绵羊膈肌样品23份,采用压片镜检法检测分离虫体并进行初步鉴定,对分离虫体的线粒体细胞色素C氧化酶第一亚基(cox1)基因部分序列(pcox1)进行PCR扩增,将测序结果与GenBank登录的其他绵羊住肉孢子虫进行序列同源性分析并构建系统发育进化树。结果显示,从23份绵羊膈肌样品中分离到11株住肉孢子虫,11株住肉孢子虫的pcox1序列长度基本一致,约为397 bp;与GenBank登录的2株柔嫩住肉孢子虫(Sarcocystis tenella)的同源性最高,分别为96.2%~99.7%和94.4%~97.9%;系统进化树分析显示,全部样品与柔嫩住肉孢子虫属同一分支,种内差异(0~5.8%)远小于种间差异(11.3%~35.5%)。研究表明,从辽宁西部地区分离到的绵羊住肉孢子虫均为柔嫩住肉孢子虫。The purpose of the experiment was to determine the species and genetic evolution of Sarcocystis in sheep in western Liaoning Province.23 sheep diaphragm samples were collected,the isolated parasites were preliminarily identified by microscopic examination,and then the partial sequence(pcox1)of the mitochondrial cytochrome C oxidase first subunit(cox1)gene of the isolated parasites was amplified by PCR.The sequence homology analysis was carried out with other Sarcocystis ovis and the phylogenetic tree was constructed.The results showed that 11 strains of Sarcocystis were isolated from 23 samples,and the pcox1 sequences of the 11 strains were basically the same in length,about 397 bp.The homology with two strains of Sarcocystis tenella registered in GenBank was the highest,which were 96.2%~99.7%and 94.4%~97.9%,respectively.And phylogenetic tree analysis showed that all samples were located in the same branch as Sarcocystis tenella,and the intraspecific differences(0~5.8%)were much smaller than the interspecific differences(11.3%~35.5%).The study indicates that all sheep Sarcocystis isolated from western Liaoning is Sarcocystis tenella.
