摘  要:目的研究峨眉山野生川黄连(野生岩连、岩连叶、雅连、味连)中盐酸小檗碱的含量测定。方法采用WondaSil C_(18) Superb色谱柱(4.6 mm×250 mm,5μm);以乙腈:0.05 mol/L磷酸二氢钾溶液(50∶50)(每100 ml中加十二烷基硫酸钠0.4 g,再以磷酸调节pH值为4.0)为流动相;检测波长为345 nm;流速:1.0 ml/min;柱温:35℃;进样量:10μl。结果盐酸小檗碱在浓度13.0258μg/ml~260.5162μg/ml范围内呈良好的线性关系,回归方程Y=44931X(R^(2)=1.00,n=5),加样回收实验的平均回收率达到101.04%(RSD=1.60%,n=6)。结论该方法简便、准确、重现性好,可用于峨眉山黄连药材中盐酸小檗碱含量的测定方法。Objective To study the determination of berberine hydrochloride in wild Coptis chinensis(Wild corydalis saxicola,Corydalis saxicola leaves,Yalian and Weilian)in Mount Emei.Method WondaSil C_(18) Superb column(4.6 mm×250 mm,5μm)was used for the determination of berberine hydrochloride in wild Coptis chinensis in Mount Emei.Acetonitrile and 0.05 mol/L potassium dihydrogen phosphate solution(50∶50)(0.4 g sodium dodecyl sμlfate was added in every 100 ml,and then the pH value was adjusted to 4.0 with phosphoric acid)were used as the mobile phase.The detection wavelength was 345 nm;The flow rate was 1.0 ml/min;Column temperature was 35℃;Sample size was 10μl.Results Berberine hydrochloride showed a good linear relationship in the range of 13.0258~260.5162μg/ml,and the regression equation was Y=44931X(R^(2)=1.00,n=5),The average recovery rate was 101.04%(RSD=1.60%,n=6).Conclusion The method is simple,accurate and reproducible,and can be used for the determination of berberine hydrochloride in wild Coptis chinensis in Mount Emei.
