摘 要:目的探讨人乳头瘤病毒16型(HPV16)E6通过微小RNA(miR)-504对宫颈癌SiHa细胞增殖、侵袭和凋亡的影响。方法将体外培养的SiHa细胞分为对照组、空载体组、E6过表达组、E6过表达+miR-NC组和E6过表达+miR-504组。采用实时荧光定量聚合酶链反应检测HPV16 E6 mRNA和miR-504的表达;采用免疫印迹法检测HPV16 E6蛋白表达;采用噻唑蓝(MTT)法、Transwell小室和流式细胞仪分别检测细胞增殖、侵袭和凋亡情况。结果与对照组和空载体组比较,E6过表达组细胞中HPV16 E6蛋白和mRNA表达水平均明显升高,而miR-504表达水平降低(P<0.05);与对照组比较,E6过表达组细胞增殖活力和侵袭能力明显升高,而细胞凋亡率降低(P<0.05);与E6过表达组和E6过表达+miR-NC组比较,E6过表达+miR-504组细胞中miR-504表达水平和细胞凋亡率明显升高,而细胞增殖活力和侵袭能力明显降低(P<0.05)。结论HPV16 E6可能通过下调miR-504的表达促进宫颈癌SiHa细胞的增殖、侵袭并抑制其细胞凋亡。Objective To investigate the effects of human papilloma virus type 16(HPV16)E6 on the proliferation,invasion and apoptosis of cervical cancer SiHa cells through microRNA(miR)-504.Methods SiHa cells were divided into control group,empty body group,E6 overexpression group,E6 overexpression+miR-NC group and E6 overexpression+miR-504 group.The expressions of HPV16 E6 mRNA and miR-504 were detected by real-time fluorescence quantitative polymerase chain reaction,the expression of HPV16 E6 was detected by Western blot,the cell proliferation,invasion and apoptosis were detected by Methyl Thiazolyl Tetrazolium(MTT),Transwell cell and flow cytometry.Results Compared with the control group or the empty body group,the expression levels of HPV16 E6 protein and mRNA in E6 overexpression group were significantly higher,while the expression level of miR-504 was significantly lower(P<0.05).Compared with the control group,the proliferative activity and invasive ability of E6 overexpression group were significantly higher,while the apoptosis rate was significantly lower(P<0.05).Compared with E6 overexpression group or E6 overexpression+miR-NC group,the expression level of miR-504 and apoptosis rate in E6 overexpression+miR-504 group were significantly higher,while the cell proliferative activity and invasive ability were significantly lower(P<0.05).Conclusions HPV16 E6 may promote proliferation,invasion and inhibit apoptosis of cervical cancer SiHa cells by down-regulating miR-504 expression.
