摘 要:目的研究矮地茶对刀豆球蛋白A(Con-A)诱导的急性免疫性肝损伤大鼠的保护作用及机制。方法将大鼠随机分为空白组、模型组及矮地茶低、中、高(10、20、40 mg·kg^(-1))剂量组,各组均通过灌胃给药;后尾静脉注射Con-A(20 mg·kg^(-1))建立肝损伤模型,测定肝脏指数,脾脏指数,血清ALT、AST、ALP水平及肝组织MDA、SOD、GSH-Px、CD4^(+)、CD8^(+)水平,HE染色观察组织病理变化。结果与空白组相比,模型组ALT、AST、ALP、MDA水平普遍升高,SOD、GSH-Px水平显著降低;与模型组相比,矮地茶低、中剂量组肝脏指数、脾脏指数,AST、ALT、ALP、MDA水平及CD4^(+)、CD8^(+)细胞数量均降低,SOD、GSH-Px升高。结论矮地茶对Con-A引起的大鼠免疫性肝损伤有一定保护作用,其机制可能与调节免疫平衡、炎症、氧化应激反应有关。Objective To determine the protective effect of Ardisiae japonicae herba on acute immunological liver injury induced by concanavalin A(Con-A)in rats and mechanism.Methods SD rats were randomly divided into a blank group,a model group,and Ardisiae japonicae herba low,middle and high dose groups(10,20,and 40 mg·kg^(-1)).Intragastric administration was used in each group.The rat model of immunological liver injury was established by tail vein injection of Con-A(20 mg·kg^(-1)).The liver-spleen coefficient,serum ALT,AST and liver tissue MDA,SOD,and GSH-Px were measured.The pathological features of hepatic tissue were observed by HE staining.The CD4^(+) and CD8^(+) positive cells in the lymphocytes were detected by ELISA.Results Compared with the blank group,the level of ALT,AST,ALP and MDA in the model group generally increased,while the level of SOD and GSH-Px in the liver tissues were decreased obviously.Compared with the model group,the liver-spleen coefficient,AST,ALT,ALP and MDA levels of rats in the low and middle dose groups of Ardisiae japonicae herba were decreased,the levels of SOD and GSH-Px in the liver tissues were increased,and the number of CD4^(+) and CD8^(+) cells in liver tissue was decreased.Conclusion Ardisiae japonicae herba has a protective effect on Con-A-induced immunological liver injury in rats,whose mechanism may be related to immunity balance regulation,inflammation and oxidative stress.
