基于高效液相色谱指纹图谱的天麻饮片-标准汤剂-配方颗粒对比研究

摘　　要：目的研究天麻饮片、标准汤剂及配方颗粒化学成分种类和量的相关性,为天麻配方颗粒的临床应用和研究提供参考。方法建立高效液相色谱指纹图谱,从化学成分种类、相似度、共有峰峰面积、指标性成分含量4个方面对天麻饮片(12批)、标准汤剂、配方颗粒(A~D厂,各3批)进行对比研究。结果饮片、标准汤剂、配方颗粒的共有峰数目分别为13个、12个、12个,与饮片(0.935)相比,标准汤剂(0.972)的相似度较高,配方颗粒(A厂0.879,B厂0.866,C厂0.854,D厂0.863)相似度稍低。以12批饮片共有峰总峰面积的均值为基准1,天麻饮片、标准汤剂及配方颗粒的相对值为饮片(1.00)、标准汤剂(0.91)、配方颗粒(A厂0.62,B厂0.47,C厂0.52,D厂0.65)。测定了天麻素、对羟基苯甲醇、巴利森苷A、巴利森苷B、巴利森苷C、巴利森苷E共6种指标成分的含量,发现与饮片相比,标准汤剂、配方颗粒中各成分含量均有不同程度降低,成分相对值顺序为天麻素>巴利森苷C>对羟基苯甲醇>巴利森苷E>巴利森苷B>巴利森苷A。结论天麻饮片、标准汤剂及配方颗粒的化学成分种类和数目基本一致,但含量存在差异,其中配方颗粒中指标成分的含量差异较明显,说明应建立全面的质量控制体系、调整临床使用当量,以促进天麻配方颗粒的临床合理应用。Objective To determine the correlation between the types and numbers of chemical components of gastrodia pieces,standard decoction and formula granules,and to provide references for the clinical application and research of gastrodia formula granules.Methods High performance liquid chromatography was used to establish a fingerprint spectrum,to campare gastrodia pieces(12 batches),standard decoctions,and formula granules(factory A-D,3 batches from each factory)from types of components,similarity,area of common peaks and index components.Results The number of common peaks were 13 in gastrodia pieces,12 in standard decoction and 12 in formula granules.Compared with the similarity of gastrodia pieces(0.935),that of the standard decoction(0.972)was a bit higher,while those of the formula granules(factory A:0.879,factory B:0.866,factory C:0.854,factory D:0.863)were lower.Based on the average value of the total peak area of 12 batches of gastrodia pieces(1),the relative values of the gastrodia pieces,standard decoction and formula granules were 1.00 for the gastrodia pieces,0.91 for the standard decoction,and for the formula granules(factory A:0.62,factory B:0.47,factory C:0.52,factory D:0.65).The content of 6 index components(gastrodin,p-hydroxybenzyl alcohol,parishin A,parishin B,parishin C,and parishin E)were determined.Compared with the gastrodia pieces,the content of each component in the formula granules and the standard decoction was reduced to different degrees.The relative value of the 6 components followed the order of gastrodin>parishin C>p-hydroxybenzyl alcohol>parishin E>parishin B>parishin A.Conclusion The types and numbers of chemical components of gastrodia pieces,standard decoction and formula granules are basically the same,but with difference in the content.The content of index components in the formula granules is significantly different,indicating that a comprehensive quality control system is needed and clinical dose should be adjusted.
