摘 要:目的建立3种半夏饮片的微生物限度检查法,通过对32批半夏的微生物限度检查,分析其微生物污染现状,为3种半夏饮片微生物的限度检查和限值的制订提供参考。方法参考《中国药典》2020年版1108中药饮片微生物限度检查法,测定3种半夏饮片中的需氧菌总数、霉菌和酵母菌总数、耐热菌总数、耐胆盐革兰氏阴性菌数,对大肠埃希菌、沙门菌进行检查,并采用全自动细菌鉴定实验系统对平板中分离得到的菌株进行鉴定。结果法半夏、姜半夏和清半夏中的需氧菌总数和霉菌酵母菌总数值均介于10~10^(5) cfu·g^(-1);耐热菌检出率为46.9%,未检出大肠埃希菌及沙门菌,但其他污染菌有成团泛菌属、非脱羧勒克氏菌、肺炎克雷伯菌肺炎、赫氏埃希菌和阴沟肠杆菌等5类。结论由于特殊的炮制工艺,半夏饮片的需氧菌总数较低,但部分样品霉菌和酵母菌总数较高,从安全性角度,建议对通过煎煮、冲泡服用的半夏应控制耐热菌并制订相关微生物限度标准。Objective To determine the microbial contamination of Chinese herbal pieces by the microbial limit test for 32 batches of Pinelliae rhizome,and to provide a reference for the microbial limit standard.Methods According to the general rules of“1108 Chinese medicine decoction pieces”in Chinese Pharmacopoeia(2020 Edition),the herbal pieces were analyzed of the total number of aerobic bacteria,yeasts,heat-resistant bacteria,bile-tolerant gram-negative bacteria,Escherichia coli,and Salmonella.The bacterial strains were identified by the automatic bacteria identification test system.Results The total aerobic bacteria and total number of molds and ranged 10~10^(5) cfu·g^(-1) in Pinelliae rhizoma praeparatum,Pinelliane rhizoma praeparatum cum zingibera et alumine,Pinelliae rhizoma praeparatum,and Pinelliae rhizoma praeparatum cum alumine.The detection rate of heat-resistant bacteria was 46.9%,while Escherichia coli and Salmonella were not detected.Other contaminated bacteria including Pantoea,Leclercia adecarboxylata,Klebsiella pneumoniae,Escherichia hermannii and Enterobacter cloacei were detected.Conclusion Due to the special processing technology,the total number of aerobic bacteria is lower in Pinelliae rhizome pieces,but the total number of molds and yeasts was higher in some samples.For safety concern,the heat-resistant bacteria should be controlled and relevant limit standards should be established for Pinelliae rhizome pieces.
