三黄紫参油治疗湿疹作用机制探讨

摘　　要：目的:探讨三黄紫参油治疗湿疹的机制。方法:从TCMSP数据库获得三黄紫参油的成分和靶点,从DisGeNET数据库获得湿疹的相关靶基因,数据处理获得交集靶点。利用STRING数据库获取交集靶点信息,使用表格展示。使用Cytoscape 3.7.2软件构建“药物-成分-靶点、疾病”拓扑网络图。用Potts-Guy模型预测药物经皮渗透性。利用DAVID 6.8在线工具对交集靶点进行GO分析和KEGG通路富集分析。结果:三黄紫参油的17种具有良好药物经皮渗透性的有效活性成分通过8个靶点发挥对湿疹的治疗作用。GO富集分析及KEGG富集分析显示,IL6、CASP8、NOS3、IL13、AHR、TP63等8个关键靶基因富集在15个生物学过程、2个细胞组分、1个分子功能、3个KEGG通路。结论:三黄紫参油通过多靶点、多通路发挥治疗湿疹的作用。作用机制可能与调控炎症应答、免疫应答,调控上皮细胞形态,调控皮肤及皮下组织细胞的细胞基本进程,调控皮肤对外部刺激的应答及局部血液循环等方面密切相关。AIM:To investigate the mechanism of the treatment of Eczema by SanHuangZiShen Oil.METHODS:The molecules and targets of SanHuangZiShen Oil were obtained from TCMSP database,the target genes of eczema were obtained from DisGeNET database,and the intersection targets were obtained by data processing.Using STRING database to obtain intersection target information,using tables to display.Cytoscape 3.7.2 software was used to construct the topological network diagram of"drug-component-target and disease".The potts-Guy model was used to predict drug transdermal permeability.GO analysis and KEGG pathway enrichment analysis were performed on the intersection targets using the DAVID 6.8 online tool..RESULTS:Seventeen active components of SanHuangZiShen Oil with good drug transdermal permeability play a therapeutic effect on eczema through 8 targets.GO enrichment analysis and KEGG enrichment analysis showed that 8 key target genes,including IL6,CASP8,NOS3,IL13,AHR and TP63,were enriched in 15 biological processes,2 cell components,1 molecular function and 3 KEGG pathways.CONCLUSION:SanHuangZiShen Oil plays a role in the treatment of eczema through multi-target and multi-pathway.The mechanism of action may be closely related to the regulation of inflammatory response,immune response,epithelial cell morphology,basic cell process of skin and subcutaneous tissue cells,skin response to external stimulation and local blood circulation.
