参地颗粒对C-BSA大鼠内质网应激及足细胞损伤的干预作用

摘  要:目的:观察中药参地颗粒对阳离子化牛血清白蛋白(C-BSA)诱导的肾炎大鼠肾脏内质网应激及足细胞自噬标志蛋白的干预作用。方法:40只雄性SD大鼠随机分为正常组、模型组、参地颗粒组、缬沙坦组,每组各10只。参地颗粒组给予参地颗粒水溶液4.0g/(kg·d)灌胃,缬沙坦组给予缬沙坦胶囊混悬液20mg/(kg·d)灌胃,正常组、模型组予生理盐水灌胃,3ml/次,1次/d,连续给药8周后处理动物、留取尿液和肾脏标本,用比色法检测24h尿蛋白含量(24hUpro),蛋白质免疫印迹法(Westernblot)检测肾脏中GRP78、CHOP、Nephrin、Podocalyxin蛋白表达。结果:与正常组比较,模型组大鼠的24hUpro显著升高(P<0.05),肾组织中GRP78、CHOP蛋白相对表达量显著增加,Nephrin、Podocalyxin蛋白相对表达量显著减少(P<0.05)。与模型组比较,参地颗粒组和缬沙坦组大鼠的24hUpro显著降低(P<0.05),肾组织中GRP78、CHOP表达量均显著减少,而Nephrin和Podocalyxin表达量则显著升高(P<0.05);且参地颗粒组GRP78、CHOP、Podocalyxin的表达变化均显著优于缬沙坦组(P<0.05)。结论:C-BSA肾炎大鼠出现内质网应激(ERS)反应增强及足细胞损伤;中药参地颗粒能够减轻内质网应激所介导的足细胞损伤,进而降低C-BSA肾炎大鼠尿蛋白水平。Objective:To observe the intervention effect of traditional Chinese medicine Shendi granule on cationized bovine serum albumin(C-BSA)-induced renal endoplasmic reticulum stress and podocyte autophagy marker protein in nephritis rats.Methods:Forty male SD rats were randomly divided into normal group,model group,Shendi granule group,and valsartan group,with 10 rats in each group.The Shendi granule group was given 4.0 g/(kg d)aqueous solution of Shendi granules by gavage,the valsartan group was given 20 mg/(kg d)of valsartan capsule suspension by gavage,and the normal group and model group were given normal saline Gavage,3ml/time,1 time/d,after continuous administration for 8 weeks,the animals were treated,urine and kidney samples were collected,the 24h urine protein content(24hUpro)was detected by colorimetry,and the western blotting method(Western blot)was used to detect GRP78,CHOP,Nephrin,Podocalyxin protein expression in kidney.Results:Compared with the normal group,the 24hUpro of the rats in the model group was significantly increased(P<0.05),the relative expressions of GRP78 and CHOP proteins in the kidney tissue were significantly increased,and the relative expressions of Nephrin and Podocalyxin proteins were significantly decreased(P<0.05).Compared with the model group,the 24hU pro of the Shendi granule group and the valsartan group was significantly decreased(P<0.05),the expressions of GRP78 and CHOP in the kidney tissue were significantly decreased,while the expressions of Nephrin and Podocalyxin were significantly increased(P<0.05);and the expression changes of GRP78,CHOP and Podocalyxin in Shendi granule group were significantly better than those in valsartan group(P<0.05).Conclusion:C-BSA nephritis rats have enhanced endoplasmic reticulum stress(ERS)response and podocyte damage;Chinese herbal medicine Shendi granule can reduce the podocyte damage mediated by endoplasmic reticulum stress,thereby reducing the severity of C-BSA nephritis of rat urine protein levels.
