摘  要:研究甾体化合物A环的C1,2-脱氢方法,并对其进行优化。采用正交设计实验方法考察了溶剂、氧气压力和催化剂对A环的C1,2-脱氢的影响。确定最佳在氧气存在条件下,乙酸做溶剂、DMSO、醋酸钯做催化剂进行脱氢效果最好,并通过底物扩展验证其适应性,制备路线具有反应条件温和,产率较高、后处理简单等优点,适合工业化生产。所有产物的结构经^(1)H NMR、^(13)C NMR确证。A new C1,2-dehydrogenation route of steroids A ring was designed and optimized.The effects of solvent,oxygen pressure,additive,catalyst on dehydrogenation were investigated by orthogonal experimental design method.The optimum process conditions were determined.Acetic acid was used as the solvent and dimethyl sulfoxide was used as the additive,the oxygen pressure was 2 MPa,and palladium acetate was used as a catalyst.Under the optimized reaction conditions,the extension of substrates to verify its suitability was performed.The preparation route had the advantages of mild reaction conditions,high yield and simple post-treatment,which was suitable for industrial production.The structures of the synthesized compounds were characterized by ^(1)H NMR,^(13)C NMR.
