摘  要:本研究旨在分析7-烟碱型乙酰胆碱受体(cholinergic receptor nicotinic alpha 7,CHRNA7)基因和酮胺还原酶晶体蛋白(m-crystallin,CRYM)基因的多态性与云上黑山羊产羔性能的关联性。利用Sequenom MassARRAY SNP技术对CHRNA7和CRYM的4个候选位点进行分型,探索其在云上黑山羊(n=544)、济宁青山羊(n=133)和辽宁绒山羊(n=91)3个群体中的遗传学特征,并对其与云上黑山羊产羔性能(包括产羔数、初生窝重和断奶窝重)的关联性进行分析。关联分析结果表明,在云上黑山羊中,CHRNA7基因的g.29127669 G>A和g.29206216 C>T两个位点及CRYM基因的g.18929951 A>G位点的各基因型对产羔数均无显著影响(P>0.05);但CRYM基因g.18929936 G>A位点AG型的产羔数显著高于GG型(P<0.05);所有候选位点的各基因型对初生窝重均无显著影响(P>0.05);CHRNA7基因g.29206216 C>T位点的CC型的断奶窝重显著高于CT和TT型(P<0.05),CRYM基因g.18929936 G>A位点的AG型的断奶窝重显著高于GG型(P<0.05)。研究结果表明,CHRNA7基因g.29206216 C>T位点可以用于断奶窝重的选择;CRYM基因的g.18929936 G>A位点可作为云上黑山羊产羔数选择的潜在分子标记,同时也适合断奶窝重的选择。This study was performed to analyze the association between the polymorphism of Cholinergic receptor nicotinic alpha 7(CHRNA7)and m-crystallin protein(CRYM)genes and kidding performance of Yunshang black goats(YS).A Sequenom Mass ARRAYSNP system was used for genotyping of the four candidate loci of CHRNA7 and CRYM,of which the genetic characteristics of YS(n=544),Jining grey goats(JN,n=133)and Liaoning cashmere goats(LN,n=91)were explored,and whose association with kidding performance including litter size,litter weight at birth and at weaning was also analyzed.The association analyses of YS showed that there were no significant effects of g.29127669 G>A and g.29206216 C>T locus at CHRNA7 and g.18929951 A>G locus at CRYM on litter size,but a significantly increased litter size of AG animals compared to GG individuals for g.18929936 G>A at CRYM(P<0.05).There were no significant effects of all loci on the litter weight at birth(P>0.05),but a striking elevated litter weight at weaning of CC animals compared to CT and TT individuals for g.29206216 C>T at CHRNA7(P<0.05),and a significantly increased litter weight at weaning of AG animals compared to GG individuals for g.18929936 G>A at CRYM(P<0.05).These results suggested that the g.29206216 C>T locus at CHRNA7was suitable for the selection on litter weight at weaning.The g.18929936 G>A locus at CRYM was the potential marker for litter size in YS,and it was suitable for selection on the litter weight at weaning.
