摘 要:本文基于UPLC-Q-Exactive-MS的代谢组学技术研究清肺化痰颗粒(QFHT)对干酵母致大鼠发热的调控作用。将SD大鼠随机分为对照组,模型组,阳性对照对乙酰氨基酚片组和QFHT高、中及低剂量组,每组10只,建立酵母诱导的大鼠发热模型,以大鼠肛温和血清中环磷酸腺苷(cAMP)的含量为指标,评价QFHT解热效果。运用UPLC-Q-Exactive-MS对大鼠血清进行非靶向代谢组学研究,获取干酵母诱导发热的潜在生物标志物,并对其进行代谢通路分析。结果表明,与模型组比较,给药组大鼠肛温和血清中c AMP含量均有所下降,有显著性差异(P<0.05);通过分析模型组和对照组血清代谢轮廓,筛选并鉴定出潜在生物标志物21个,主要涉及两条代谢途径。QFHT对发热大鼠的治疗作用与其改善血清中潜在生物标志物的水平有关,且主要通过调控色氨酸代谢、牛磺酸和亚牛磺酸代谢途径发挥解热作用。To study the regulation effect of Qingfei Huatan granule(QFHT)on the dry yeast induced fever in rats based on the UPLC-Q-Exactive-MS metabonomics technique.SD rats were divided into the control group,model group,positive control acetaminophen tablet group and QFHT high,medium and low dose group randomly,with 10 rats in each group.A yeast induced rat fever model was established,and the antipyretic effect of QFHT was evaluated by the content of c AMP in rat anus and serum.UPLC-Q-Exactive-MS was used to conduct untargeted metabolomics studies on rat serum to obtain potential biomarkers of dry yeast-induced fever and analyze their metabolic pathways.The results showed that compared with the model group,c AMP content in anal and serum of rats in administration group was decreased,with significant difference(P<0.05).By analyzing the serum metabolic profiles of model group and control group,21 potential biomarkers were screened and identified,mainly involving two metabolic pathways.The therapeutic effect of QFHT on febrile rats is related to the improvement of the level of potential biomarkers in serum,and the antipyretic effect of QFHT is mainly through regulating the metabolism of tryptophan,taurine and taurine.
