摘 要:黄酮类化合物是酸枣仁中的主要生物活性成分,具有镇静、催眠和强心等功效。为了研究酸枣仁中黄酮类化合物的种类和数量,利用超高效液相色谱电喷雾离子源三重四极杆串联质谱法(UPLC/ESI-Q TRAP-MS/MS)对酸枣仁中的黄酮类化合物进行分离鉴定分析。采用Waters ACQUITY UPLC HSS T3 C18色谱柱,流动相为超纯水(0.04%的乙酸)乙腈(0.04%的乙酸)。在电喷雾离子源(electrospray ionization,ESI)和三重四级杆(QQQ)检测模式下对酸枣仁中的黄酮类化合物进行扫描检测。利用Analyst 1.6.3软件处理质谱数据,基于MWDB数据库,对获得的代谢物质谱进行数据分析。结果显示基于UPLC/ESI-Q TRAP-MS/MS技术从酸枣仁中共分离鉴定出83个黄酮类化合物,包括3个查尔酮类、5个二氢黄酮类、2个二氢黄酮醇类、5个花青素类、18个黄酮类、22个黄酮碳糖苷类、14个黄酮醇、9个黄烷醇类和5个异黄酮类。本研究可丰富酸枣仁中黄酮类化合物的种类和数量,为酸枣仁的药效成分和质量控制研究奠定基础。Flavonoids are the main bioactive components in Ziziphi spinosae Semen,which have sedative,hypnotic and heart-strengthening effects.In order to study the types and quantities of the flavonoids in Ziziphi spinosae Semen,the flavonoids in Ziziphi spinosae Semen wereanalyzed and identified using the UPLC/ESI-Q TRAP-MS/MS.The separation was performed on a Waters ACQUITY UPLC HSS T3C18 column with mobile phases of water(0.04%acetic acid)and acetonitrile(0.04%acetic acid).The extraction of flavonoids compounds was evaluated under electrospray ionization(ESI)and triple quadrupole(QQQ)in Ziziphi spinosae Semen.The mass spectrometry data were processed by Analyst 1.6.3 software,and the obtained metabolite spectra were analyzed based on MWDB database.The results showed that83 flavonoids compounds were identified from Ziziphi spinosae Semen in this study,including 3 chalcones,5 dihydroflavonoids,2 dihydroflavonols,5 anthocyanins,18 flavonoids,22 flavonoid carboglycosides,14 flavonols,9 flavanols and 5 isoflavones.This study enriched the variety and quantity of flavonoids compounds in Ziziphi spinosae Semen,which would provide a base for the study of pharmacodynamic components and quality control of Ziziphi spinosae Semen.
