摘  要:钙(Ca)是植物必需的一种大量元素,可作为信号分子转导多种生理过程,钙的缺失或过量均会影响植物生长发育。桑树是重要的多年生经济树木,目前对桑树中microRNA(miRNA)的抗性功能和胁迫响应调控的研究较少。使用桑树品种育71-1为研究材料,构建小RNA(small RNA)文库。通过小RNA深度测序,鉴定得到53个已知miRNA和42个预测的新型miRNA成熟体。3个钙胁迫比较组合中的差异表达miRNA共36个,其中19个上调和17个下调。挑选miR156g为研究对象,预测其茎环结构,符合二级结构规则:成熟体位于5′端,前体自由能为-166.523 kJ/mol,具有典型茎环结构;通过psTarget软件预测到对应靶标mRNA大部分属于SPL家族。试验结果为桑树钙胁迫调控的分子机制以及miRNA功能的研究提供了依据。Calcium(Ca)is an essential and abundant element in plants,which can be used as a signaling molecule to transduce various physiological processes.The lack or excess of Ca will affect the growth and development of plants.Mullberry is an important economic perennial tree.However,there are few studies on the resistance function of micro-RNAs in mulberry and the regulation of stress response.In this study,Yu 71-1 was used as research material to construct a small RNA library.By deep sequencing of the small RNA,53 known miRNAs and 42 predicted mature novel miRNAs were identified.A total of 36 miRNAs were differentially expressed in the 3 calcium stress comparison combinations,including 19 up-regulated and 17 down-regulated miRNAs.The stem-loop structure of miR156 g was predicted,which was in accordance with the secondary structure rules.The mature body was located at the 5′end,and the free energy of the precursor was-166.523 kJ/mol,showing a typical stem-loop structure.Most of the corresponding target mRNAs were predicted to belong to the SPL family by psTarget software.This study provided a basis for the study on the molecular mechanism of calcium stress regulation and miRNA function in mulberry.
