摘 要:目的:基于“肾-唾、脾-涎”理论,考察肾虚质、脾虚质大鼠唾液腺肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、γ-干扰素(interferon-γ,IFN-γ)、白细胞介素-10(interleukin-10,IL-10)的表达差异。方法:采用“猫吓鼠”造模法制备先天不足+后天失养复合型肾虚质模型,采用饮食不节+劳倦过度法制备脾虚质模型,正常孕鼠所产仔鼠为平和质组。模型建立完成后,比较各组仔鼠的体质量;收集唾液,检测各组仔鼠唾液pH值;ELISA法检测唾液腺中TNF-α、IFN-γ、IL-10的含量。结果:与平和质组比较,肾虚质组仔鼠第8周体质量显著降低(P<0.01),脾虚质组仔鼠第6、8周体质量均显著降低(P<0.01);肾虚质组、脾虚质组仔鼠唾液pH值显著升高(P<0.01),唾液腺中IFN-γ、IL-10的含量显著降低(P<0.01);脾虚质组仔鼠唾液腺中TNF-α含量显著升高(P<0.05)。与肾虚质组比较,脾虚质组第6、8周时仔鼠体质量均显著降低(P<0.01),但唾液pH值及唾液腺中TNF-α、IFN-γ、IL-10含量无明显变化。结论:肾虚质、脾虚质组大鼠免疫稳态失衡,但二者组间差异不明显,提示唾液为脾肾共同所主。Objective:Based on the theory of"Kidney-saliva and Spleen-salivation",to investigate the salivary gland tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)and interleukin-10(IL-10).Methods:The"cat scaring mouse"model was used to prepare the complex model of congenital deficiency+acquired dystrophy,and the model of spleen deficiency was established by the method of poor diet+overwork.After the model was established,the body weights of the mice in each group were compared;saliva was collected to detect the pH value of the saliva of the mice in each group;the contents of TNF-α,IFN-γand IL-10 in salivary glands were detected by ELISA.Results:Compared with the Ping He constitution group,the body weight of the mice in the Kidney-deficiency group decreased significantly at the 8th week(P<0.01),and the body weight of the Spleen-deficiency group decreased significantly at the 6th and 8th week(P<0.01).The salivary pH value of mice in Spleen deficiency group was significantly increased(P<0.01),and the contents of IFN-γand IL-10 in salivary glands were significantly decreased(P<0.01);the content of TNF-αin salivary glands of mice in Spleen deficiency group significantly increased High(P<0.05).Compared with the Kidney-deficiency group,the body weight of the Spleen-deficiency group was significantly decreased at the 6th and 8th week(P<0.01),but the pH value of saliva and the contents of TNF-α,IFN-γ,and IL-10 in salivary glands had no significant changes.Conclusion:Compared with the normal group,the growth and development,salivary pH value,salivary gland TNF-α,IFN-γ,and IL-10 expressions of the rats in the Kidney-deficiency and Spleen-deficiency groups were significantly different from those in the normal group,but the difference between the two groups was not significant,suggesting that the saliva is affected by both Spleen and Kidney.
