摘 要:目的研究西红花苷对心肌缺血再灌注损伤糖酵解的干预作用及糖酵解相关酶的影响。方法采用H9c2大鼠心肌细胞株建立缺氧/复氧体外心肌缺血再灌输损伤模型,通过MTT实验评价西红花苷对H9c2模型细胞增殖的影响,检测H9c2细胞中葡萄糖消耗,乳酸含量评价西红花苷对H9c2模型细胞糖酵解的影响;通过检测ATP及NAD+/NADH水平,评价西红花苷对细胞能量供应水平的影响;检测己糖激酶2活性及蛋白表达,考察西红花苷对糖酵解相关酶活性影响及蛋白含量的影响,明确其对缺氧/复氧细胞中己糖激酶2的调控作用。结果西红花苷对缺氧/复氧H9c2细胞增殖具有明显促进作用,并呈浓度依赖性;西红花苷可以明显升高模型细胞中葡萄糖消耗及乳酸含量,并促进细胞ATP产生,降低NAD+/NADH含量,促进肿瘤细胞糖酵解水平。西红花苷可升高己糖激酶Ⅱ活性,上调其蛋白表达水平。当给于己糖激酶抑制剂后,西红花苷对模型细胞糖酵解调控作用及己糖激酶Ⅱ调节作用明显降低,但与抑制剂组仍存在显著差异。结论西红花苷可以明显促进缺氧/复氧心肌细胞增殖,并升高其糖酵解水平及其能量供应,同时明显升高心肌细胞中己糖激酶Ⅱ的表达。Objective To investigate the the intervention effects of crocin on glycolysis and glycolysis related enzymes in myocardial ischemia-reperfusion injury.Methods The rat cardiomyocyte line(H9c2)was used to establish the model of hypoxia/reoxygenation myocardial ischemia in vitro.The effects of crocin on the proliferation of H9c2 model cells were detected by MTT assay,and the levels of glucose consumption and lactic acid were measured to evaluate the effects of crocin on the glycolysis of H9c2 model cells.The effects of crocin on cell energy supply were evaluated by detecting the levels of ATP and NAD+/NADH.Moreover the activity and protein expression levels of hexokinase 2 were detected to investigate the effects of crocin on the activity of glycolysis related enzymes and protein content,and to clarify its regulatory effects on hexokinase 2 in hypoxia/reoxygenation cells.Results The crocin could significantly promote the proliferation of hypoxia/reoxygenation H9c2 cells in a concentration dependent manner,which could significantly increase the glucose consumption and lactic acid content in model cells,promote the production of ATP,reduce the content of NAD+/NADH,and promote the glycolysis level of tumor cells.In addition,crocin could increase the activity of hexokinaseⅡand up regulate its protein expression levels.After administration of hexokinase inhibitor,the effects of crocin on glycolysis and hexokinase 2 of model cells were decreased significantly,but there were still significant differences between crocin group and inhibitor group.Conclusion The crocin can significantly promote the proliferation of hypoxic/reoxygenated cardiomyocytes,increase the glycolysis level and energy supply,moreover,which can significantly increase the expression levels of hexokinase 2 in cardiomyocytes.
