荷叶总生物碱的制备方法研究

摘　　要：目的:建立大孔吸附树脂-分散固相萃取法的荷叶总生物碱的制备方法。方法:采用乙醇加热回流法从荷叶中提取生物碱,然后采用大孔吸附树脂法对荷叶提取物进行纯化,再采用PCX分散固相萃取技术对大孔吸附树脂纯化物进一步纯化,最终得荷叶总生物碱纯化物。结果:采用85%乙醇为提取溶剂对荷叶中生物碱进行提取,然后采用D101大孔吸附树脂对荷叶醇提物进行纯化,收集1~3 BV的60%和70%乙醇,得大孔吸附树脂纯化物,此时树脂纯化物中荷叶碱的含量为6.84%~7.40%。最后,以PCX为吸附剂,采用分散固相萃取技术进一步进行纯化,以5%氨-甲醇进行洗脱,回收溶剂得到荷叶总生物碱纯化物,其中纯化物中荷叶碱的含量为27.66%~30.12%。结论:该方法经过大孔吸附树脂、分散固相萃取两步纯化,所得荷叶生物碱纯化物中荷叶碱含量较高。Objective:To establish a method for the preparation of total alkaloids from Nelumbinis Folium by macroporous adsorption resin and dispersive solid phase extraction.Methods:The alkaloids were extracted from Nelumbinis Folium by ethanol heating reflux,and the purification process of macroporous adsorption resin was adopted to remove the impurities.After then,the PCX dispersed solid-phase extraction was used and the total alkaloids was obtained.Results:The alkaloids in Nelumbinis Folium was extracted with 85%ethanol as the extraction solvent,and then the ethanol extract from Nelumbinis Folium was purified with D101 macroporous adsorption resin.60%and 70%ethanol of 1-3 BV were collected to obtain macroporous adsorption resin purification.At this time,the content of nuciferinea in the resin purification was between 6.84%-7.40%.Finally,PCX was used as adsorbent,further purified by dispersive solid-phase extraction technology,elution was conducted with 5%ammonia-methanol,and the total alkaloids purified by recovered solvent was obtained,among which,the content of nuciferinea in the purified product was 27.66%-30.12%.Conclusion:The purification of total alkaloids from Nelumbinis Folium is preferable by macroporous adsorption resin and solid phase extraction,and the purificated product has high content of nuciferine.
